摘 要:2C型蛋白磷酸酶(PP2C)是最大的蛋白磷酸酶家族,PP2C蛋白在响应不同非生物胁迫中发挥着重要作用。为了探究人参Pg PP2C2基因的功能,基于实验室前期转录组数据库,从人参中克隆了一个2C型蛋白磷酸酶基因,命名为Pg PP2C2,并对Pg PP2C2基因的序列、编码蛋白质的结构进行分析,预测了其功能。研究结果表明,该核苷酸序列的开放阅读框为1269 bp,编码422个氨基酸。其编码蛋白质属于亲水性蛋白,无信号肽,未形成跨膜结构域,包含2C家族丝氨酸/苏氨酸磷酸酶催化结构域。通过蛋白互作分析,发现Pg PP2C2蛋白与HOG1(参与渗透调节的丝裂原活化蛋白激酶)和NBP2(NAP1-binding蛋白2,参与HOG途径的蛋白)的互作分数分别高达0.996和0.984,推测Pg PP2C2蛋白可能参与渗透胁迫调节及磷酸化过程。本研究结果可为后期进一步研究PgPP2C2基因的功能和开展人参种植调控提供理论基础。Type 2C protein phosphatases(PP2C)are the largest family of protein phosphatases,which plays an important role in responding to different abiotic stresses.In order to explore the function of Panax ginseng PgPP2C2 gene,based on the transcriptome database in the early stage of the laboratory,a 2C protein phosphatase gene was cloned from ginseng,named PgPP2C2,and the sequence and protein structure of PgPP2C2 gene were analyzed,and its function was predicted.The results of the study showed that the nucleotide sequence obtained an open reading frame of 1269 bp,encoding 422 amino acids.The encoded protein,a hydrophilic protein with no signal peptide,no transmembrane domain,contained the serine/threonine phosphatase catalytic domain of 2C family.Through protein interaction analysis,it was found that the interaction scores of PgPP2C2 protein with HOG1(mitogen-activated protein kinase involved in osmotic regulation)and NBP2(NAP1-binding protein 2,a protein involved in the HOG pathway)were as high as 0.996 and 0.984,respectively.It is speculated that PgPP2C2 protein may be involved in the regulation of osmotic stress and the process of phosphorylation.The results of this study can provide a theoretical basis for further research on the function of the PgPP2C2 gene and the regulation of Panax ginseng planting.
