摘 要:蔗糖转运蛋白(SUTs)在介导蔗糖由源到库器官的长距离运输、植物生长发育以及抵抗逆境胁迫中发挥重要作用,研究白芨蔗糖转运蛋白BsSUT2的序列信息与表达模式,为揭示其蛋白结构及基因功能提供依据。基于白芨转录组数据,采用RT-PCR技术获得白芨Bs SUT2基因CDS全长,通过生物信息学软件分析Bs SUT2编码蛋白的分子特征,并对BsSUT2进行氨基酸序列比对与系统进化树分析;采用实时荧光定量PCR技术检测BsSUT2基因的组织表达模式。结果表明,克隆获得的BsSUT2基因CDS全长为1584 bp,编码527个氨基酸。BsSUT2蛋白包含12个跨膜结构域,亚细胞定位预测表明BsSUT2蛋白定位于质膜,与铁皮石斛和小兰屿蝴蝶兰SUT2基因编码蛋白序列一致性高,隶属于SUT4亚族。Bs SUT2基因的表达具有组织特异性,其转录本在白芨幼苗的假鳞茎中表达量最高。推测BsSUT2基因在白芨生长发育和蔗糖的分配中发挥重要作用。本研究对白芨BsSUT2基因的序列特征与组织表达模式进行分析,为阐明Bs SUT2基因在蔗糖运输与分配、生长发育和逆境胁迫中的功能提供实验数据支持。SUTs plays an important role in mediating the long-distance transportation of sucrose from source to sink organs,plant growth,and resistance to stress.The sequence information and expression pattern of sucrose transporter Bs SUT2 in Bletilla striata were studied to provide the basis for revealing its protein structure and gene function.Based on the transcriptome data of Bletilla striata,the full length CDS of BsSUT2 gene was obtained by RT-PCR,the molecular characteristics of BsSUT2 were analyzed by bioinformatics tools.Amino acid sequence alignment and phylogenetic analysis of BsSUT2 were performed.The quantitative real-time PCR(RT-qPCR)was performed to investigate the expression patterns of BsSUT2.The result showed that the BsSUT2 cDNA contained an open reading frame of 1584 bp nucleotides encoding a 527 amino acids protein.It was predicated that the protein exhibited 12 transmembrane spanning domain.Subcellular localization prediction indicated that BsSUT2 protein was localized in the plasma membrane.The phylogenetic comparisons revealed that BsSUT2 was a member of SUT4 clade and shared high identity with DeSUC2 in Dendrobium catenatum and PeSUC 2 in Phalaenopsis equestris.The expression of BsSUT2 revealed differential expression by tissue specificity,and showed much higher expression in pseudobulb of seeding.Our findings suggested that Bs SUT2 gene may play an important role during plant growth and the distribution of sucrose.The analysis of sequence and tissue expression pattern of BssUT2 gene provided experimental data for elucidating the function of BsSUT2 gene in sucrose transport and distribution,growth and development,and adversity stress responses.
