摘 要:为建立甜瓜杂交种纯度快速鉴定的方法,以杂交新品种‘川蜜脆玉’及其亲本为试验材料,利用改良的胚根DNA速提法和SSR(Simple sequence repeats)分子标记技术检测种子纯度及真实性,并结合大田的形态学鉴定结果进行验证。结果表明,在18对SSR引物中,有7对引物在‘川蜜脆玉’F1代及父母本中呈现多态性。其中SSR00398与MU4104-1引物特异性好、条带清晰、父母本条带间隔明显,作为纯度鉴定的引物,鉴定纯度为98.1%,与田间鉴定结合符合率在99%以上,说明这2对引物能够作为‘川蜜脆玉’杂交种纯度鉴定的特异引物进行纯度检测。新方法的建立大大缩短了甜瓜杂交种的鉴定周期,提高鉴定效率,为甜瓜新品种的杂交种快速鉴定提供重要技术支撑。In order to establish a rapid purity identification method of hybrid melon seed with SSR markers,new melon hybrid‘Chuanmicuiyu’and its parental lines as materials.The improved radical DNA extraction method and simple sequence repeat(SSR)molecular markers were used to test variety purity and authenticity identification of‘Chuanmicuiyu’,and the results were verified by field morphological identification.The results showed that there were seven pairs of primers in the SSR primers of 18 pairs,which had a good polymorphism between the F generation of‘Chuanmicuiyu’and its parents.Among them,SSR00398 and MU4104-1 primers had good specificity,clear bands and obvious band spacing,which were the primers for the purity identification of melon.The identification results were 98.1%.Compared with results of morphological identification of‘Chuanmicuiyu’in field,the coincidence rate of the variety purity detected by SSR markers were higher than 99%.SSR00398 and MU4104-1 could be used as the specific primer to identify the purity of hybrid-‘Chuanmicuiyu’.The establishment of the new method greatly shortened the identification cycle of melon hybrids and greatly improved the identification efficiency,which provided important technical support for rapid identification of new varieties of melon hybrids.
