摘  要:目的优化4种儿茶素类物质及咖啡因的加热浸提工艺,系统性地研究上述组分在茶叶中不同叶位的分布差异,为茶叶的生产、加工提供理论指导。方法采用单因素和L_(9)(3^(4))正交实验对表没食子儿茶素没食子酸酯(EGCG)、表儿茶素没食子酸酯(ECG)、表没食子儿茶素(EGC)、儿茶素[(+)-catechin,C]和咖啡因(CAF)的提取条件进行工艺优化,获得最优组合。以高效液相色谱法检测凤冈锌硒茶福鼎大白茶树品种不同叶位中上述5种成分的含量。结果建立了最优的加热浸提工艺:温度90℃,乙醇浓度为60%,料液比1∶40,第1次提取时间35 min,第2次提取时间10 min。影响提取含量的因素依次为:温度>料液比>提取液浓度>提取时间。获得了4种儿茶素类物质及咖啡因在凤冈锌硒茶不同叶位的含量,其分布规律如下:EGCG和总儿茶素含量均为一叶>二叶>芽>三叶>成叶>茎,ECG为芽>一叶>二叶>三叶>茎>成叶,CAF为一叶>芽>二叶>三叶>茎>成叶,EGC为一叶>二叶>三叶>芽>茎>成叶,C为芽>茎>一叶>二叶>三叶>成叶。结论优化后的提取工艺简单、稳定、重复性好,可用于茶叶中儿茶素及咖啡因的检测。凤冈锌硒茶中4种儿茶素类成分及咖啡因含量在茶树嫩梢不同部位有一定差异。4种儿茶素类及咖啡因的含量均随叶位排序负相关,即叶片成熟度越高上述成分的含量越低。本研究可为凤冈锌硒茶儿茶素类物质和咖啡因的提取及含量测定提供参考,并为茶叶的采摘及充分利用提供理论基础。Objective To optimize the heating extraction process of four catechins and caffeine contents in Fenggang zinc selenium tea,and systematically study the distribution differences of these active components in different positions of the tea leaves,so as to provide theoretical guidance for the production and processing of functional tea.Methods Using single factor and L 9(3^(4))orthogonal test,the extraction conditions of epigallocatechin gallate(EGCG),epigallocatechin(EGC),epicatechin gallate(ECG),(+)-catechin(C)and caffeine(CAF)were optimized and the optimal combination was obtained.The contents of these five components in different positions of the Fenggang zinc selenium tea leaves were determined by high performance liquid chromatography.Results The optimal extraction process established was as follows:heating at 90℃,ethanol concentration 60%,solid-liquid ratio 1∶40,the first extraction lasted for 35 min plus the second extraction lasted for 10 min.The factors that affect the extraction content in sequence are:temperature>solid-liquid ratio>extraction concentration>extraction time.The contents of four catechins and caffeine in different leaf positions of Fenggang-zinc-selenium tea were obtained.The distribution of EGCG content and total content of catechins were as follows:1 st leaf>2 nd leaf>bud>3 rd leaf>mature leaf>stem,while the distribution of ECG content was:bud>1 st leaf>2 nd leaf>3 rd leaf>stem>mature leaf,CAF content as:1 st leaf>bud>2 nd leaf>3 rd leaf>stem>mature leaf,EGC content as:1 st leaf>2 nd leaf>3 rd leaf>bud>stem>mature leaf,and the C content as:bud>stem>1 st leaf>2 nd leaf>3 rd leaf>mature leaf.Conclusion The optimized extraction process is simple,stable and reproducible,which can be used for the detection of catechins and caffeine in tea.The contents of 4 catechins and caffeine showed certain difference among different parts of tender shoot in Fenggang zinc Selenium tea.Their content was negatively correlated with the order of leaf positions,as the higher the leaf maturity was,the lower t
