摘 要:目的:研究心衰康胶囊对糖尿病大鼠心肌纤维化的改善作用,并初步判断其机制。方法:选用清洁级、雄性SD大鼠32只,随机取8只为对照组,余大鼠以1%链脲佐菌素(STZ)溶液经尾静脉注射制备糖尿病模型,造模成功者随机分为模型组、心衰康胶囊组、缬沙坦组,每组8只。对照组和模型组均给予生理盐水灌胃;心衰康胶囊组和缬沙坦组分别给药灌胃,定期监测血糖、体质量,10W治疗结束后检测大鼠左室射血分数(EF)、短轴缩短率(FS),计算心脏及左室指数,后处死大鼠取左心室。通过苏木素-伊红(HE)染色和Masson染色观察心肌组织病理形态表现,采用免疫组织化学法检测转化生长因子-β1(TGF-β1)蛋白表达水平。结果:与对照组比较,模型组血糖水平、TGF-β1水平均显著升高(P<0.05,P<0.01);体质量、心脏指数、左室指数、EF及FS均显著降低(P<0.05,P<0.01);HE:心肌结构破坏明显;Masson:胶原沉积明显;免疫组织化学法:心肌纤维化明显,TGF-β1蛋白表达明显增强。与模型组比较:药物组血糖无明显下降(P>0.05),差异无统计学意义;余指标均有不同程度逆转(P<0.05,P<0.01)。结论:心衰康胶囊可通过抑制TGF-β1蛋白表达,延缓糖尿病大鼠心肌纤维化进程,改善心功能。Objective:To study the improvement effect of Xinshuaikang capsule on myocardial fibrosis in diabetic rats and to preliminarily judge its mechanism.Methods:32male SD Rats of clean grade were selected,and 8rats were randomly selected as the control group.The remaining rats were injected with 1%Streptozotocin(STZ)solution through the tail vein to prepare the diabetes model.Those who succeeded in modeling were randomly divided into the model group,the Xinshuaikang capsule group,and the Valsartan group,with 8rats in each group.Both the control group and the model group were given physiological saline by gavage;The Xinshuaikang capsule group and the Valsartan group were administered by gavage respectively,and the blood sugar and body weight were monitored regularly.After 10W treatment,the left ventricular Ejection fraction(EF)and short axis shortening rate(FS)of rats were measured,and the heart and left ventricular index were calculated.The rats were killed and left ventricles were taken.The pathological morphology of myocardial tissue was observed by hematoxylin eosin(HE)staining and Masson staining,and Transforming growth factor was detected by immunohistochemistry-β1(TGF-β1)Protein expression level.Results:Compared with the control group,the blood glucose level and TGF-β1All levels significantly increased(P<0.05,P<0.01);Body mass,cardiac index,left ventricular index,EF,and FS were significantly reduced(P<0.05,P<0.01);HE:Significant damage to myocardial structure;Masson:Significant collagen deposition;Immunohistochemical method:Significant myocardial fibrosis,TGF-β1protein was significantly enhanced.Compared with the model group,there was no significant decrease in blood glucose in the drug group(P>0.05),and the difference was not statistically significant;All other indicators showed varying degrees of reversal(P<0.05,P<0.01).Conclusion:Xinshuaikang capsules can inhibit TGF-β1protein expression,delay the process of myocardial fibrosis in diabetes rats,and improve cardiac function.
