摘 要:目的:通过研究粪菌移植干预,进一步验证麸炒苍术发挥抗脾虚治疗作用与肠道菌群密切相关。方法:将SPF级小鼠随机分为空白组(C组)、脾虚模型组(M组)、脾虚造模+移植脾虚大鼠粪菌组(FMTm组)、脾虚造模+移植正常大鼠粪菌组(FMTz组)、脾虚造模+移植麸炒苍术粪菌组(FMTf组),每组10只。除空白组外,其余各组小鼠均采用番泻叶灌胃加饥饱失常复制小鼠脾虚模型,观察并记录小鼠状况,以出现泄下纳呆、毛色枯槁、蜷缩聚堆和反应迟钝等表现即为造模成功。实验第8天开始各造模组分别灌胃相应粪菌液,连续7天。灌胃结束后,采用酶联免疫吸附法(ELISA)测定各组小鼠血清中胃泌素(GAS)、胃动素(MTL)、血管活性肠肽(VIP)、淀粉酶(AMS)水平。结果:与空白组比较,模型组小鼠血清中GAS、MTL、AMS和VIP含量均显著升高(P<0.05),胃肠激素紊乱,与模型组比较,经粪菌移植后FMTz组和FMTf组各指标含量均显著降低(P<0.05),脾虚症状明显改善。结论:移植正常大鼠粪菌和移植麸炒苍术组大鼠粪菌后均对脾虚小鼠有明显改善作用,表明麸炒苍术可通过调节肠道菌群紊乱而发挥健脾作用。Objective:This paper will further investigate whether the anti-spleen deficiency depends on intestinal flora.Methods:SPF mice were randomly divided into blank groups(C),spleen deficiency model group(M),spleen deficiency+transplanted rats(FMTm),spleen deficiency+transplanted normal rats(FMTz)and 10spleen deficiency+gluten-fried feces bacteria(FMTf).Except for the blank group,all mice used the spleen deficiency model of gagage lavage and satiety replication mice to observe and record the condition in order to show the performance of diarrhea,withered hair color,curled pile and slow reaction.From the eighth day of the experiment,each module was infused with the corresponding fecal fluid for 7consecutive days.After the completion of gavage,the serum levels of gastrin(GAS),gastrin(MTL),vasoactive intestinal peptide(VIP),and amylase(AMS)were determined by an enzyme-linked immunosorbent assay(ELISA).Results:GAS,MTL,AMS and VIP increased(P<0.05),gastrointestinal hormone disorder,and compared with the model group,FMTz and FMTf were significantly decreased(P<0.05)and spleen deficiency symptoms improved.Conclusion:Both normal mice and bran-fried miceimproved the spleen deficiency mice,indicating that they strengthened the spleen by regulating intestinal flora.
