儿童清咽解热口服液体外抗呼吸道合胞病毒(RSV)作用及其抗炎活性研究

摘　　要：目的:研究儿童清咽解热口服液体外抗呼吸道合胞病毒(RSV)作用及其抗炎活性。方法:以利巴韦林(Ribavirin)作为阳性对照药物,在HEP-2细胞水平上,采用CellTiter-GloTM免疫荧光法测定受试样品的细胞毒性,通过间接免疫荧光法测定流感病毒蛋白表达水平,以检测细胞培养液上清中病毒复制水平,评价儿童清咽解热口服液对呼吸道合胞病毒(RSV)的作用,同时采用AlphaLISA法检测培养上清中IL-6、IL-8、MCP-1等炎症因子的表达水平。结果:儿童清咽解热口服液半数中毒浓度(CC_(50))为15.33mg·mL^(-1),对呼吸道合胞病毒(RSV)的半数有效浓度(EC_(50))为1.27mg·mL^(-1),炎性指标IL-6、IL-8、MCP-1的半数有效浓度(EC_(50))分别为0.73mg·mL^(-1)、0.83mg·mL^(-1)和0.61mg·mL^(-1)。呼吸道合胞病毒(RSV)选择指数(SI)为12.07,炎性指标IL-6、IL-8、MCP-1的SI分别为21、18.5和25.3。结论:在HEP-2细胞水平上,儿童清咽解热口服液对呼吸道合胞病毒(RSV)病毒具有明显的抗病毒作用,并显示出良好的抗炎活性。Objective:The aim of this study is to test the anti-virus effect and anti-inflammatory activity of the children Qingyan Jiere oral liquid to respiratory syncytial virus(RSV)in cellular level.Methods:The anti-virus effect of children Qingyan Jiere oral liquid to RSV was evaluated,at the HEP-2cell level,ribavirin was used as a positive control drug.While,the inflammatory factors level such as IL-6,IL-8,MCP-1was detected in the culture supernatant in the way of alphalisa method.The cytotoxicity of the tested samples was determined by CellTiter-GloTMimmunofluorescence method and the expression level of influenza virus protein was determined by indirect immunofluorescence method to detect the virus replication level in the supernatant of cell culture medium.Results:The concentration of cytotoxicity 50%(CC_(50))of children Qingyan Jiere oral liquid was 15.33mg·mL^(-1),the concentration for 50%of maximal effect(EC_(50))against respiratory syncytial virus(RSV)was 1.27mg·mL^(-1),and the EC_(50)of inflammatory indicators IL-6,IL-8and MCP-1were 0.73mg·mL^(-1),0.83mg·mL^(-1)and 0.61mg·mL^(-1)respectively.The selecion index(SI)of RSV was 12.07,and the SIof IL-6,IL-8and MCP-1were 21,18.5and 25.3,respectively.Conclusion:At HEP-2cells level,children Qingyan Jiere oral liquid has obvious antiviral effect on respiratory syncytial virus(RSV),and shows good anti-inflammatory activity.
