摘  要:本文以木犀草苷为对照品,采用紫外-可见分光光度计法建立了一种黄芫花中总黄酮含量测定方法,并对黄芫花中总黄酮的提取工艺进行了优化。结果表明木犀草苷在质量浓度为4.1μg/mL~20.3μg/mL范围内与吸光度的线性关系良好,回归曲线方程为A=48.967 C-0.010 8,R=0.999 9(n=5),且重复性(RSD=0.29%,n=6)和精密性(RSD=0.33%,n=6)良好,平均回收率为97.86%(RSD=2.08%,n=9)。L_(9)(3^(4))正交试验优化黄芫花总黄酮提取工艺结果显示,超声提取法提取黄芫花总黄酮的最佳提取工艺为:乙醇体积分数60%、超声时间0.5 h、提取功率160 W、料液比1∶20(g/mL)。最后对5个不同产地黄芫花总黄酮含量进行了测定,结果显示,山西绛县黄芫花总黄酮含量最高为22.81 mg/g,来自山东舜王城的黄芫花总黄酮含量最低为18.08 mg/g。以上结果表明,本文所建立的黄芫花总黄酮含量测定方法和提取工艺流程,操作简捷易行,提取效率高,稳定性好,适合于黄芫花总黄酮的提取和含量测定。In this paper, using cynaroside as the control a method of the content determination of total flavonoids from the buds of W.chamaedaphne by UV-Vis spectrophotometry was established. Meanwhile the extraction process of total flavonoids from the buds of W. chamaedaphne was optimized. The results showed that the linearity of cynaroside was good at the mass concentration range of 4.1-20.3 μg/mL with absorbance, the regression curve equation were A=48.967 C-0.010 8,R=0.999 9(n=5), and the reproducibility(RSD=0.29%, n=6) and precision(RSD=0.33%, n=6) were good with a mean recovery of 97.86%(RSD = 2.08%, n=9). The results of L_(9)(3^(4)) orthogonal test to optimize the extraction process of total flavonoids from the buds of W. chamaedaphne showed that the best extraction process was extracted by ultrasonic with 60% ethanol, 0.5 h extraction time, 160 W extraction power, and 1∶20(g/mL)material to liquid ratio. Finally, the total flavonoids content of the buds of W. chamaedaphne collected from five different origins was determined. The results showed that the highest total flavonoids content was 22.81 mg/g in Jiangxian County, Shanxi Province, and the lowest total flavonoids content was 18.08 mg/g from Shunwangcheng Village, Shandong Province. The above results showed that the method for the determination of total flavonoids from the buds of W. chamaedaphne and the extraction process established in this paper is simple and easy to operate, with high extraction efficiency and good stability, and are suitable for the extraction and determination of total flavonoids from the buds of W. chamaedaphne.
