摘 要:为有效提高大花杓兰地下芽的成活率,优化其生长条件,以大花杓兰花期、果期和休眠期的地下芽为材料,在单因素试验的基础上,通过L9(34)正交试验对组织培养方法进行优化,探索培养基、6-BA、NAA和pH对大花杓兰地下芽成活率的影响。结果表明:休眠期微剥鳞叶芽成活率优于花期和果期的成活率,最高达84.44%;地下芽成活率影响因素主次顺序为培养基>6-BA>NAA>pH;大花杓兰地下芽的最佳培养基为MS+1.0 mg/L 6-BA+0.1 mg/L NAA+pH 6,其成活率高达73.33%,且地下芽生长状态良好,苗株健康。优化后的组织培养方法能显著提高大花杓兰地下芽成活率,为大花杓兰苗株大规模培养提供理论参考。To effectively improve the survival rate and growth conditions of the underground buds of Cypripedium macranthos Sw,the tissue culture method was optimized by L9(34)orthogonal test based on a single-factor experiment,and the effects of medium,6-BA,NAA,and pH on the survival rate of underground buds of C.macranthos Sw were explored.The results showed that the survival rate of micro-scaly leaf buds in the dormant stage was better than that at the flowering and fruit stages,up to 84.44%.The primary and secondary factors influencing the survival rate of underground buds were the medium>6-BA>NAA>pH.The best medium for underground buds of C.macranthos Sw was MS+1.0 mg·L^(–1)6-BA+0.1 mg·L^(–1)NAA+pH 6,and the survival rate was as high as 73.33%,the underground buds were in good condition and the seedlings were healthy.The optimized tissue culture method can significantly improve its survival rate,which provides a theoretical reference for the large-scale cultivation of plants of C.macranthos Sw.
