摘 要:目的:探讨鸢尾素(irisin)在运动改善去势大鼠骨质疏松中的作用机制。方法:将100只雌性SD大鼠(清洁级)随机分为空白组、模型组、假手术组、运动组和模型+鸢尾素(irisin)注射组,每组各20只。空白组不做任何治疗,模型组、运动组和模型+irisin注射组行双侧卵巢摘除术造大鼠骨质疏松症模型。假手术组只切除少量肠系膜,保留卵巢,不做任何治疗。模型鉴定成功后,运动组进行运动训练,模型+irisin注射组行双侧卵巢摘除术造大鼠骨质疏松症模型,每天一次irisin肌肉注射,空白组、模型组和假手术组不做任何治疗。治疗后测定大鼠腰椎和股骨骨密度(BMD),血清irisin、成骨细胞特异性转录因子2(Runx2)、骨保护素(OPG)表达水平,碱性磷酸酶(ALP)、Runx2及OPG蛋白表达水平,Runx2、OPG基因表达含量,骨髓中骨钙素(OC)、I型前胶原氨基肽(PINP)含量。结果:与模型组相比,运动组和模型+irisin注射组腰椎及股骨BMD、血清irisin、Runx2、OPG含量、ALP、Runx2、OPG蛋白表达量、Runx2、OPG mRNA表达水平、骨髓中OC和PINP含量均有所提高(P<0.05)。结论:运动可有效增加去势大鼠的骨密度,其机制可能是运动能通过irisin增加ALP活性,促进BMSCs增殖,上调Runx2、OPG mRNA表达,对成骨及破骨双重调节,进而达到防治骨质疏松的目的。Objective:To explore the mechanism of action of irisin in improving ovariectomized osteoporosis rats through exercise.Method:100 female SD rats(clean grade)were randomLy divided into blank group,model group,sham operation group,exercise group,and model+irisin injection group,with 20 rats in each group.The blank group did not receive any treatment,while the model group,exercise group,and model+irisin injection group underwent double ovariectomy to create a rat model of osteoporosis.The sham surgery group only removed a small amount of mesentery and retained the ovaries without any treatment.After successful model identification,the exercise group underwent exercise training,while the model+irisin injection group underwent double ovariectomy to create a rat osteoporosis model.Irisin was intramuscularly injected once a day,while the blank group,model group,and sham operation group did not receive any treatment.After treatment,bone mineral density(BMD)of the lumbar spine and femur of rats,serum levels of irisin,osteoblast specific transcription factor 2(Runx2),osteoprotegerin(OPG)expression,alkaline phosphatase(ALP),Runx2,and OPG protein expression levels,Runx2,OPG gene expression levels,and osteocalcin(OC)and type I procollagen aminopeptide(PINP)levels in bone marrow were measured.Result:Compared with the model group,the exercise group and the model+irisin injection group showed significant increases in lumbar and femoral BMD,serum irisin,Runx2,OPG content,ALP,Runx2,OPG protein expression,Runx2,OPG mRNA expression levels,and bone marrow OC and PINP content(P<0.05).Conclusion:Exercise can effectively increase bone density in ovariectomized rats,and its mechanism may be that exercise can increase ALP activity through irisin,promote BMSCs proliferation,upregulate Runx2 and OPG mRNA expression,and regulate both osteogenesis and osteoclasis,thereby achieving the goal of preventing and treating osteoporosis.
