摘 要:单分子测序技术是一种无需聚合酶链式反应扩增即可对每个核酸分子单独测序的技术。它具有读长长、通量高、测序速度快等优点,是核酸测序技术史上一次重大的革新。单分子实时测序和纳米孔单分子测序是单分子测序技术中的两个典型代表。本文通过总结这两者的特点、原理及测序的基本过程,概述单分子测序技术在粮食作物、经济林木、药用植物等基因组和转录组研究领域的进展,以期为农、林相关植物分子生物学相关研究提供参考。Single-molecule sequencing is a technology in which each nucleic acid molecule can be individually sequenced without polymerase chain reaction amplification.It has the advantages of long reading length,high throughput,high sequencing speed,and is a major innovation in the history of nucleic acid sequencing technology.Single-molecule real-time sequencing and nanopore single molecule sequencing are two typical representatives of single-molecule sequencing technology.In this paper,the characteristics,principles and basic process of sequencing of these two technologies were summarized,and the progress of single-molecule sequencing technology in genome and transcriptome research of food crops,economic trees and medicinal plants were summarized,in order to provide reference for the research on molecular biology of agriculture and forestry related plants.
