摘 要:目的:探究孕期“恐伤肾”对仔鼠肠道菌群结构的影响,并预测其关键菌群及差异代谢功能。方法:基线一致的已孕0.5 d Wistar大鼠分为正常组和恐应激组,两组于孕18 d采用悬尾实验、糖偏好实验评价造模效果。仔鼠延续母鼠分组,在5周龄行悬尾试验、高架实验评价仔鼠情绪,使用16S rRNA基因测序分析仔鼠肠道菌群。结果:与正常组比较,恐应激组孕鼠悬尾不动时间显著增加(P<0.05),糖偏好率显著降低(P<0.05);恐应激组仔鼠悬尾不动时间显著增加(P<0.05),高架开放臂停留时间比例显著减少(P<0.05)。两组仔鼠肠道菌群丰富度、多样性、物种结构差异显著(P<0.05)。与正常组比较,恐应激组厚壁菌门(Firmicutes)显著降低,拟杆菌门(Bacteroidetes)显著升高(LDA>4.0,P<0.05),菌属方面乳杆菌属(Lactobacillus)、粪杆菌属(Faecalibacterium)、嗜果胶拟杆菌属(Bacteroides pectinophilus)显著降低(LDA>4.0,P<0.05),瘤胃球菌UCG014(Ruminococcaceae UCG014)、Erysipelotrichaceae科的杜波氏菌属(Dubosiella)、异芽杆菌属(Allobaculum)、尿道杆菌属(Turicibacter)显著升高(LDA>4.0,P<0.05)。菌群代谢功能方面,与正常组比较,恐应激组仔鼠外源生物降解与代谢、脂质代谢功能丰度降低(P<0.05),能量代谢功能丰度增加(P<0.05)。结论:孕期恐应激导致仔鼠不良情绪,伴随肠道菌群紊乱与其代谢功能的失调。Objective:To investigate the structural changes of intestinal flora and predict the key flora and differential metabolic functions in the offspring rats by‘fear impairing kidney’during pregnancy.Methods:Wistar rats at 0.5 days of gestation at baseline were divided into normal and fear-stress groups,and the modeling effect was evaluated by tail suspension test and sugar preference test at 18 days of gestation in both groups.Their offspring continued to be grouped with their dams,and the mood of offspring rats were evaluated by tail suspension test and elevation test at 5 weeks of age.The gut microbiota of the offspring rats was analyzed using 16S rRNA gene sequencing.Results:Compared with the normal group,the pregnant rats in the fear-stress group showed a significant increase in tail suspension immobility time(P<0.05)and a significant decrease in sugar preference rate(P<0.05).The offspring rats in the fear-stress group showed a significant increase in tail suspension immobility time(P<0.05)and a significant decrease in the proportion of time spent in the open arm of the elevation(P<0.05).The differences in intestinal flora richness,diversity,and species structure between the two groups of offspring rats were significant(P<0.05).Compared with the normal group,the phylum Firmicutes was significantly lower and the phylum Bacteroidetes was significantly higher(LDA>4.0,P<0.05)in the fear-stress group,and the genera Lactobacillus,Faecalibacterium,and Bacteroides pectinophilus(P<0.05)were significantly lower.Bacteroides pectinophilus were significantly lower(LDA>4.0,P<0.05),and Ruminococcaceae UCG014,Dubosiella,Allobaculum,and Turicibacter of Erysipelotrichaceae were significantly elevated(LDA>4.0,P<0.05).Aspects of the flora metabolic function,the abundance of exogenous biodegradation and metabolism and lipid metabolism functions were reduced(P<0.05)and energy metabolism functions were increased(P<0.05)in the littermates of the fear-stress group compared to the normal group.Conclusion:Pregnancy fear-stress leads to
