摘 要:目的:运用含药血清经时谱效关系研究经典名方半夏白术天麻汤保护内皮细胞免受棕榈酸诱导细胞焦亡的药效物质。方法:建立棕榈酸诱导的人脐静脉内皮细胞(HUVEC)焦亡模型,以细胞活力、一氧化氮(NO)、丙二醛(MDA)、总超氧化物歧化酶(T-SOD),抑制其释放炎性介质白介素-6(IL-6)、肿瘤坏死因子α(TNF-α)作为半夏白术天麻汤含药血清抑制细胞焦亡的药效学评价指标,建立大鼠灌胃后吸收进入血化学成分的分析方法,采用典型相关性的谱效关系分析方法指认半夏白术天麻汤抑制细胞焦亡的有效成分。结果:柚皮苷、大豆苷元、甘草素、异甘草素和白术内酯Ⅲ与抑制细胞焦亡的正相关性较大,可明显增强细胞活力,改善MDA、NO、T-SOD、IL-6、TNF-α水平。结论:半夏白术天麻汤中橘红、甘草、白术的成分参与保护棕榈酸诱导的HUVEC细胞焦亡,含药血清经时谱效关系可用于发现半夏白术天麻汤的血管内皮细胞保护成分。Objective:To study the pharmacodynamic effect of the classic prescription Banxia Baizhu Tianma Decoction(BBTD)to protect endothelial cells pyroptosis induced by palmitic acid by using the time-dependent spectrum-effect relationship of serum containing drugs.Methods:Human umbilical vein endothelial cells(HUVEC)pyroptosis model induced by palmitic acid was established.Using cell viability,nitric oxide(NO),malondialdehyde(MDA),total activity of superoxide dismutase(T-SOD),interleukin(IL)-6 and tumor necrosis factor-α(TNF-α)as pharmacodynamic evaluation indexes for inhibiting pyroptosis in drug-containing serum,a method for analyzing chemical components absorbed into blood was established after gavage in rats,and the effective components of inhibiting pyroptosis in BBTD were identified by time-dependent spectrum-effect analysis of typical correlation.Results:The positive correlation of Naringin,Daidzein,Liquiritigenin,Isoliquiritigenin and AtractylenolideⅢwas significant,which could obviously enhance the cell viability and improve the levels of MDA,NO,T-SOD,IL-6 and TNF-α.Conclusion:The ingredients in Citri Exocarpium Rubrum,Glycyrrhizae Radix et Rhizoma,Atractylodis Macrocephalae Rhizoma can protect HUVEC cell induced by palmitic acid,and the time-dependent spectrum-effect relationship of serum containing drugs can be used to find the protective components of vascular endothelial cells in BBTD.
