摘  要:目的:观察电针内关、公孙穴对功能性消化不良(FD)大鼠抑郁样行为、内脏敏感性和下丘脑促肾上腺皮质激素释放因子(CRF)及其受体CRF1、CRF2蛋白表达的影响。方法:将40只SPF级SD大鼠随机分为空白组、模型组、电针组、西药组,每组10只,采用复合病因造模法制造FD模型,造模成功后电针组大鼠行电针干预,穴取双侧内关、公孙穴,疏密波,频率为(2/10)Hz,电流强度为1 mA,每次30 min;西药组灌服莫沙必利和黛力新溶液;均每日干预1次,连续21 d。干预结束后采用旷场实验观察焦虑抑郁行为;腹壁撤退反射评分(AWR)评定内脏敏感性;Western Blot法检测下丘脑组织CRF及其受体CRF1、CRF2蛋白表达。结果:与空白组比较,模型组大鼠在旷场箱中的穿格、起立、修饰次数显著减少(P<0.05);AWR评分在球囊内压力为40、60、80 mmHg时显著升高(P<0.05);下丘脑CRF及其受体CRF1蛋白表达显著上调,CRF2受体蛋白表达显著下调(P<0.05)。与模型组比较,电针组、西药组大鼠在旷场箱中的穿格、起立、修饰次数显著增加(P<0.05);AWR评分在球囊内压力为40、60、80 mmHg时评分显著降低(P<0.05);下丘脑CRF及其受体CRF1蛋白表达显著下调,CRF2蛋白表达显著上调(P<0.05)。与西药组比较,电针组各项指标差异均无统计学意义。结论:电针内关、公孙穴可明显改善FD大鼠焦虑抑郁行为和内脏高敏性,其作用机制可能与下调下丘脑CRF及其受体CRF1蛋白表达,上调CRF2受体蛋白表达,进而调节CRF1-CRF2受体信号平衡通路有关。Objective:To observe the effects of electroacupuncture at Neiguan(PC 6)and Gongsun(SP 4)acupoints on depression like behavior,visceral sensitivity and the protein expression of hypothalamic CRF and its receptors CRF1 and CRF2 in rats with functional dyspepsia(FD),and to explore the possible mechanism of electroacupuncture at Neiguan(PC 6)and Gongsun(SP 4)acupoints in the treatment of FD.Methods:Forty SPF SD rats were randomly divided into blank group(10 rats),model group(10 rats),electroacupuncture group(10 rats)and western medicine group(10 rats),and the FD model was created by compound etiology modeling method.After the successful modeling,the rats in the electroacupuncture group were subjected to electroacupuncture intervention,with acupoints Neiguan(PC 6)and Gongsun(SP 4)on both sides,sparse and dense waves,frequency(2/10)Hz,current intensity 1 mA,for 30 min each time;the western medicine group was given mosapride and dalexin solution;both interventions were performed once a day for 21 d.After the intervention,the anxiety and depression behaviors were observed by the open field experiment;the visceral sensitivity was assessed by the abdominal wall retreat reflex score(AWR);the Western Blot method was used to detect the hypothalamic tissue expression of CRF and its receptors CRF1 and CRF2.Results:Compared with the blank group,the times of grid piercing,standing up and modification in the open field box of the model group were significantly reduced(P<0.05);the AWR score increased significantly when the balloon pressure was 40,60 and 80 mmHg(P<0.05);the expression of CRF and its receptor CRF1 protein in hypothalamus increased and the expression of CRF2 receptor protein significantly decreased(P<0.05).Compared with the model group,the times of grid piercing,standing up and modification in the open field box of rats in electroacupuncture group and western medicine group were significantly increased(P<0.05);the AWR score decreased significantly when the balloon pressure was 40,60 and 80 mmHg(P<0.05);the expression
