摘  要:目前,黑土退化问题日益严重,有机肥添加是改善黑土农田肥力退化的有效措施之一,但有机肥中抗生细菌或抗生素抗性基因存在污染土壤的风险。为了探讨畜禽粪便有机肥对黑土农田中抗生素抗性基因的影响,研究采用荧光定量分析的方法,探讨施用无机肥和不同种类有机肥(包括牛粪、猪粪和鸡粪)条件下,黑土农田土壤中抗生素抗性基因数量和丰度变化。结果表明,施用猪粪和鸡粪有机肥土壤中抗性基因种类较多,四环素类是研究中主要的抗性基因类型,检出比例高达55%以上。相对于施用无机肥的土壤,施用牛粪有机肥土壤抗性基因特征无明显变异,施用猪粪有机肥增加了土壤中β-内酰胺类抗生素抗性基因blaTEM丰度,而施用鸡粪有机肥降低了blaTEM等抗性基因丰度,显著增加了tetX1等四环素类抗生素抗性基因丰度。总的来说,畜禽粪便有机肥施用加剧了土壤中抗生素抗性基因的污染,且不同有机肥施用形成了土壤中不同的抗性基因分布格局。研究结果将从黑土农田土壤中抗生素抗性基因积累角度,为合理制定有机肥施肥制度、保障生态安全和人类健康管理提供理论依据。At present,the degradation of black soil is becoming more and more serious,application of organic fertilizer is widely considered as one of the effective measures to improve soil fertility.However,antibiotic-resistant bacteria or antibiotic resistance genes in organic fertilizer may cause soil pollution.In order to study the effect of livestock manure organic fertilizer on antibiotic resistance genes in black soil farmland,quantitative PCR analysis was used to investigate the changes in the number and abundance of antibiotic resistance genes in black soil farmland under the conditions of inorganic fertilizer and different types of organic fertilizer(including cow manure,pig manure and chicken manure).The results showed that the antibiotic resistance genes detected in soil fertilized with pig manure and chicken manure were more than that with cow manure,and the tetracyclines was the major antibiotic resistance gene type in this study with high detection ratio of more than 55%.Compared with inorganic fertilizer application,cow manure application did not significantly change the distribution characteristics of resistance genes in soil,pig manure application increased the blaTEM abundance ofβ-lactam antibiotic resistance genes,while chicken manure application significantly decreased the abundance of gene blaTEM and increased the tetX1 abundance of tetracyclines resistance genes.Overall,livestock manure organic fertilizer application increased the contamination of antibiotic resistance genes in soil,and different distribution patterns of antibiotic resistance genes were formed in soil applied with different types of organic fertilizers.From the perspective of antibiotic resistance gene accumulation in black soil farmland,the results will provide a theoretical basis for development of organic fertilizer fertilization system,ecological security and human health management.
