摘 要:为探索蒲公英中黄酮类化合物的最佳提取条件,以乙醇为溶剂,采用超声波辅助法对蒲公英植株和蒲公英花中黄酮类化合物的提取进行了研究。选择料液比、超声时间、温度三个单因素进行试验以初步探寻黄酮类化合物的提取条件,根据单因素试验结果进行正交试验设计确定最佳提取条件。利用可见光谱法和红外光谱法对蒲公英粗提液中的黄酮类化合物进行了定性和定量分析。结果表明:蒲公英植株中黄酮类化合物的最佳提取条件为:料液比1∶50(g∶mL),超声时间40 min,温度60℃;蒲公英花中黄酮类化合物的最佳提取条件为:料液比1∶40(g∶mL),超声时间50 min,温度70℃。蒲公英植株和蒲公英花中粗提液中黄酮类化合物的得率分别为4.86%和3.95%,这为蒲公英植株及花中黄酮类化合物的进一步研究及其合理利用奠定一定的基础。To explore the optimal extraction conditions for flavonoids in dandelion,the extraction of flavonoids from dandelion plants and dandelion flowers was investigated by ultrasonic assisted method using ethanol as solvent.Three single factors,solid-liquid ratio,ultrasonic time and temperature,were selected to explore the extraction conditions of flavonoids.According to the single factor test results,orthogonal experiment design was carried out to determine the best extraction conditions.The qualitative and quantitative analysis of the flavonoids in the crude extract of Dandelion was carried out by Vis and IR spectroscopy.The results showed that the best extraction conditions for flavonoid compounds in dandelion plants were:solid-liquid ratio of 1∶50(g∶mL),sonication time of 40 min and sonication temperature of 60℃;the best extraction conditions for flavonoid compounds in dandelion flowers were:solid-liquid ratio 1∶40(g∶mL),ultrasonic time 50 min,temperature 70℃.The extraction rates of flavonoids in the crude extracts from dandelion plants and dandelion flowers were 4.86%and 3.95%,respectively,which laid a certain foundation for further research on flavonoids in dandelion plants and flowers and their rational utilization.
