摘  要:植物特有的YABBY转录因子家族在植物生长发育、激素信号响应、抗逆性、作物育种和农业生产等方面发挥重要作用。为了探究陆地棉与海岛棉YABBY转录因子的特征,本研究在陆地棉和海岛棉中共鉴定出49个YABBY基因,其中陆地棉25个,海岛棉24个,其编码的氨基酸的序列长度在119~221个氨基酸之间,均为亲水性蛋白,所有成员都定位在细胞核中。二级结构含量较高的元件是无规则卷曲。根据基因结构和蛋白保守基序的相似性以及系统进化树将其分为5个亚族,分别为YAB2、YAB5、CRC、FIL/YAB3和INO。共线性分析表明YABBY在陆地棉和海岛棉种间同源染色体之间呈现很好的共线性,海岛棉基因Gbar_A06G005560.1在陆地棉中有Ghir_A06G005750.1和Ghir_A06G005760.1两个拷贝。基因表达模式分析发现,陆地棉与海岛棉之间YABBY同源基因在棉花不同组织和不同发育阶段表达存在差异,例如在陆地棉中Ghir_A05G001610.1在纤维发育第10天高量表达,而在海岛棉中其同源基因Gbar_A05G001620.1在纤维发育第10天不表达,陆地棉Ghir_A04G004440.1在纤维发育第5天高量表达,其海岛棉中同源基因Gbar_A04G003930.1在纤维发育第10天、20天和30天均高表达,这些基因可能导致不同棉种间棉纤维发育的差异。本研究结果为进一步研究棉花YABBY基因的功能,尤其是在纤维发育方面奠定了理论基础。The plant-specific YABBY transcription factor family plays an important role in plant growth and development,plant hormone signal response,stress resistance,crop breeding and agricultural production.In order to explore the difference of YABBY transcription factors between Gossypium hirsutum and Gossypium barbadense,a total of 49 YABBY genes were identified in this study,including 25 in G.hirsutum and 24 in G.barbadense.The amino acid sequences encoded by YABBY genes ranged from 119 to 221 amino acids,all of which were hydrophilic proteins,and all members were located in the nucleus.The element with higher secondary structure content is random coil.According to the similarity of gene structure and protein conserved motif and phylogenetic tree,they are divided into 5 sub-families,namely YAB2,YAB5,CRC,FIL/YAB3 and INO.Collinearity analysis showed that YABBY exhibits good collinearity between homologous chromosomes of G.hirsutum and G.barbadense.The gene Gbar_A06G005560.1 of G.barbadense has two copies of Ghir_A06G005750.1 and Ghir_A06G005760.1 in upland cotton.Analysis of gene expression patterns revealed that the YABBY homologous gene expression level between G.hirsutum and G.barbadense is different in different tissues and different developmental stages of cotton.For example,in upland cotton,the gene Ghir_A05G001610.1 was expressed at a high level on the 10thday of fiber development,while its homologous gene Gbar_A05G001620.1 was not expressed at the same time in G.barbadense.The gene Ghir_A04G004440.1 was highly expressed on the 5thday of fiber development in G.hirsutum,while its homologous gene Gbar_A04G003930.1 in the G.barbadense was highly expressed on the 10th,20thand30thday of fiber development.These genes may cause differences in cotton fiber development among different cotton species.These results lay a theoretical foundation for further research on the function of cotton YABBY gene,especially in fiber development.
