摘 要:为研究光敏色素作用因子(Phytochrome-interacting factor,PIF)基因在白菜(Brassica rapa)生长发育及逆境胁迫中的功能,利用生物信息学和转录组学的方法鉴定了白菜PIF基因家族成员并分析了这些家族成员的蛋白质理化性质、基因结构、染色体分布、系统发育进化关系、启动子顺式作用元件以及表达模式。结果表明:白菜中含有10个BrPIF基因,分布于6条染色体上,含有10个保守基序和6~8个外显子;BrPIF蛋白呈酸性或碱性,均定位于细胞核上;10个基因根据系统进化树可分为3个簇;顺式作用元件分析得到BrPIF家族成员含有光响应、低温、激素等响应元件,且大部分白菜PIF家族基因在经过温度胁迫处理后转录水平发生显著变化,荧光定量分析表明这些基因在不同的组织部位表达量也各有高低,表明这些基因参与温度等非生物逆境胁迫并在不同部位的生长发育中发挥了作用。本研究为白菜PIF基因功能的挖掘提供科学依据。To study the function of phytochrome-interacting factor(PIF)gene in the growth and development and the adversity stress response of cabbage(Brassica rapa),the BrPIF gene family members were identified and analyzed by bioinformatics and transcriptomics.The protein physicochemical properties,gene structure,evolutionary relationship,chromosome distribution,promoter cis-acting elements and expression patterns of BrPIFs were analyzed.The results showed that there were ten BrPIFs was distributed on six chromosomes,containing ten conserved motifs and 6~8 exons.BrPIF protein was acidic or alkaline,and located on the nucleus.According to the phylogenetic tree,BrPIFs can be divided into three clusters.The cis-acting element analysis showed that the BrPIFs involved in light response,low temperature,and hormone.The transcription level of the BrPIFs changed significantly in after temperature stress.The expression levels of these genes were different in different tissues.They were involved in abiotic stress and played a role in the growth and development of different parts.This research provided a foundation for further study the roles of PIF in B.rapa.
