摘  要:胶孢炭疽菌(Colletotrichum gloeosporioides)复合种下果生刺盘孢(C.fructicola)引起的草莓炭疽病严重危害草莓产业,病原菌通过分泌效应蛋白协助侵染宿主。为了研究果生刺盘孢效应蛋白与草莓互作机理,本研究通过生物信息学对草莓果生刺盘孢的效应蛋白进行了全基因组预测,并对所得候选蛋白进行序列分析。结果表明,共获得355个候选效应蛋白,分属于29个已知蛋白家族;转录组测序结果显示260个候选效应蛋白在侵染草莓后的24、72和96 h均显著上调表达,60个蛋白在三个时间点均下调表达;对候选效应蛋白进行了蛋白互作网络分析,获得两个蛋白互作模块,发现其参与宿主细胞壁裂解和调控菌株致病力。本研究为探究草莓炭疽病的致病机理研究提供了分子靶标。Strawberry anthracnose caused by Colletotrichum fructicola(C.fructicola)of the species complex(C.gloeosporioides)sensu lato,seriously endangers the strawberry industry.The pathogen intends to infect the host by secreting multiple effector proteins.In order to clarify the protein interaction mechanism between C.gloeosporioides and strawberry,genome-wide prediction of effector proteins in C.gloeosporioides was conducted by bioinformatics,and the obtained candidate sequences were further screened and analyzed.In total 355 candidate effectors were finally harvested,which belonged to 29 known protein families.Transcriptomic sequencing results showed that the expression of 260 candidate effector proteins were among them were significantly up-regulated at 24,72 and 96 h post strawberry inoculation,and 60 proteins were down-regulated at these three time points.The candidate effector proteins were analyzed by protein interaction network,and two protein interaction modules were obtained,and it was found that they were involved in host cell wall lysis and regulation of strain pathogenicity.This research provided crucial molecular targets for exploring the pathogenesis of strawberry anthracnose.
