摘  要:为了解析垂柳丛枝病发病机理,本研究以健康垂柳和感丛枝病垂柳为材料,采用转录组测序技术,检测和分析两者之间差异基因的表达定量、功能注释、富集通路及候选基因。结果表明:共检测到差异基因18536个,上调和下调表达基因分别为10316个、8220个。GO功能分析显示代谢过程和有机物代谢过程在生物学过程中被注释得最多;在细胞组分本体中,被注释得最多次数的条目是细胞器、蛋白复合物和细胞内细胞器;注释到分子功能本体中最多的条目为催化活性分子和氧化还原酶活性。通过KEGG富集分析结果,筛选出光合作用、植物-病原相互作用、苯丙氨酸、酪氨酸和色氨酸生物合成、光合作用-天线蛋白、卟啉与叶绿素代谢、亚油酸、氨基糖和核苷酸糖代谢和α-亚麻酸代谢共8条通路与垂柳丛枝病发生密切相关,并筛选出K02183(CALM)、K13424(WRKY33)、K13448(CML)、K13459(RPS2)、K13457(RPS3)、K13412(CPK)6个垂柳丛枝病发生相关的候选基因。本研究结果为揭示垂柳丛枝病发生的分子机制提供一定的参考。In order to study the pathogenesis of weeping willow witches'broom disease,we used healthy weeping willows and weeping willows infected with witches'broom as materials and adopted the technology of transcriptome sequencing,the differential gene expression,functional annotation,enrichment pathway and candidate genes were detected and analyzed.The results showed that the 18536 differential genes were detected,with 10316 upregulated and 8220 down-regulated.GO function analysis showed that metabolic processes and organic metabolism processes were most annotated in biological processes;In the cell component,the most frequently annotated entries are organelles,protein complexes,and intracellular organelles;The most frequently annotated entries in the functional ontology of molecules are catalytic active molecules and oxidoreductase activities.Through KEGG enrichment analysis results,8 pathways of are closely related to the occurrence of weeping willow witches'broom disease of photosynthesis,plant pathogen interaction,phenylalanine,tyrosine and tryptophan biosynthesis,photosynthesis antenna protein,porphyrin and chlorophyll metabolism,linoleic acid,amino sugar and nucleotide sugar metabolism andα-linolenic acid metabolism.Six candidate genes related to the occurrence of weeping willow witches'broom disease,namely K02183(CALM),K13424(WRKY33),K13448(CML),K13459(RPS2),K13457(RPS3),and K13412(CPK),have been screened.The results have important reference value for further revealing the molecular mechanism of weeping willow witches'broom disease.
