摘  要:利用筛选的37对多态性SSR引物对天津科润黄瓜研究所育成的16个黄瓜主栽品种进行SSR-DNA扩增,并构建黄瓜品种SSR特征指纹图谱,以鉴定各品种的真实性和一致性。结果表明:37对引物共扩增出94个位点,其中多态性位点78个,多态性比例为82.98%。引物SSR13466、SSR10018和SSR21164区分能力表现最强,可将16个品种分别区分为6、5和4个类型。利用其中的24对引物构建的指纹图谱可有效区分所有16个供试品种,联合应用其中的2~3对引物可以将16个品种中每一个品种进行准确区分,除‘津优35’、‘津优301’和‘津优409’外,其余13个品种均有1~5个单标记即可与其他15个品种区分。16个黄瓜品种间的遗传相似系数在0.5638~0.9787之间,采用UPGMA法进行聚类,在遗传相似系数为0.67处将16个品种聚为2类,聚类与品种栽培茬口和育种部门无明显的相关性。该结果可作为16个黄瓜主栽品种真实性、纯度等鉴定的重要参考依据,对黄瓜种子的质量管理以及知识产权保护具有重要意义。37 pairs of polymorphic SSR primers were used to amplify SSR-DNA of 16 main cucumber varieties bred by Tianjin Kernel Cucumber Research Institute,and the SSR fingerprint of cucumber varieties was constructed to identify the authenticity and consistency of each variety.The results showed that 94 loci were amplified by37 pairs of primers,including 78 polymorphic loci,and the proportion of polymorphism was 82.98%.SSR13466,SSR10018 and SSR21164 had the strongest distinguishing ability,which could divide 16 varieties into 6,5 and 4types respectively.The fingerprint constructed by using 24 pairs of primers can effectively distinguish all 16 tested varieties,each of the 16 varieties can be completely distinguished by using 2~3 pairs of primers in 24 pairs of polymorphic core primers.Except for'Jinyou35','J inyou301'and'Jinyou409',the other 13 varieties can be distinguished from other 15 varieties with 1~5 single markers.The genetic similarity coefficient of 16 cucumber varieties ranged from 0.5638 to 0.9787,Which was clustered by UPGMA method,and the 16 varieties were grouped into two groups at the genetic similarity coefficient of 0.67,and there was no obvious correlation between clustering and variety cultivation stubble,breeding departments.The results can be used as an important reference for the identification of authenticity and purity of 16 main cucumber varieties,which is of great significance to the quality management and intellectual property protection of cucumber seeds.
