摘  要:为了筛选出合适的抗生素种类和浓度作为遗传转化体系的抗性筛选剂,本研究以卡那霉素、遗传霉素(G418)、潮霉素B、草甘膦作为筛选剂,对椰枣愈伤组织进行敏感性试验。以椰枣茎尖生长组织为外植体诱导出的愈伤组织为实验材料,分别设计卡那霉素、遗传霉素(G418)、潮霉素B、草甘膦4种筛选剂不同浓度梯度,研究负筛选剂卡那霉素、遗传霉素(G418)、潮霉素B、草甘膦对椰枣胚性愈伤组织的致死浓度、半致死浓度及时间,确定愈伤组织对筛选剂的敏感性,为建立椰枣负筛选系统的遗传转化体系提供基础。结果表明:(1)椰枣愈伤组织对筛选剂的敏感度:G418>潮霉素B>卡那霉素>草甘膦。(2)50 mg/L的G418浓度适合作为椰枣遗传转化体系中的抗性筛选剂。(3)50 mg/L的潮霉素B浓度适合作为椰枣遗传转化体系中的抗性筛选剂。(4)100 mg/L的卡那霉素浓度适合作为椰枣遗传转化体系中的抗性筛选剂。(5)椰枣对草甘膦不敏感,不适用作为遗传转化的抗性筛选剂。综上所述,G418和潮霉素B更适合作为椰枣遗传转化体系过程中的抗性筛选剂。In order to select suitable antibiotic types and concentrations as resistant screening agents for genetic transformation system,kanamycin,genomycin(G418),hygromycin B and glyphosate were used as screening agents to perform sensitivity tests on callus of date palm.Callus induced from shoot tip of date palm was used as experimental material,individually designed gradient,to study the sensitivity of embryogenic callus to negative screening reagent kanamycin,genamycin(G418),hygromycin B and glyphosate in induction stage,in order to establish date palm negative screening system foundation for genetic transformation system.The results showed that:(1)The sensitivity of callus to screening reagent was G418hygromycin Bkanamycinglyphosate.(2)50 mg/L G418 was suitable as a resistance screening agent in the genetic transformation system of date palm.(3)Hygromycin B concentration of 50 mg/L was suitable as a resistance screening reagent in the genetic transformation system of date palm.(4)The concentration of kanamycin 100 mg/L was suitable as a resistance screening reagent in the genetic transformation system of date palm.(5)Date palm is not sensitive to glyphosate and is not suitable as a resistance screening reagent for genetic transformation.In summary,G418 and hygromycin B are more suitable as resistance screening agents in the process of genetic transformation system of date palm.
