左归丸对5/6肾切除大鼠IL-1β、p65及胸主动脉钙化的影响

摘　　要：目的:观察左归丸对5/6肾切除大鼠IL-1β、p65及胸主动脉钙化的影响。方法:SPF级Wistar雄性大鼠予以5/6肾切除并给予高磷饮食造模,造模大鼠分为模型组、左归丸组,并同时设空白对照组。术后空白对照组予以普通饮水,模型组和左归丸组予以高磷饮水。术后4周空白对照组和模型组给予灭菌注射用水灌胃,左归丸组行左归丸水溶液灌胃,共8周。灌胃8周后处死大鼠,检测血清BUN、Scr、IL-1β,免疫组化检测胸主动脉IL-1β、p65表达,von kossa染色观察胸主动脉钙化情况。结果:给药8周后,空白对照组和左归丸组BUN与模型组比较,差异有统计学意义(P<0.05)。空白对照组和左归丸组Scr与模型组比较,差异有统计学意义(分别为P<0.01,0.05)。空白对照组和左归丸组IL-1β与模型组比较,差异有统计学意义(P<0.01)。空白对照组大鼠各组IL-1β、p65在动脉血管壁几乎无表达,模型组大鼠表达明显,左归丸组大鼠与空白对照组比较增加,但明显少于模型组;von kossa染色观察空白对照组大鼠胸主动脉中内膜层无明显钙盐沉积,模型组大鼠主动脉钙盐沉积明显,左归丸组钙盐沉积较模型组明显减轻。结论:中药左归丸能够改善5/6肾切除大鼠胸主动脉钙化,其机制可能与抑制IL-1β、p65炎性因子表达有关。Objective:To observe the effect of Zuogui pill on IL-1β,p65 and aortic calcification in 5/6 nephrectomy rats.Methods:SPF grade male Wistar rats were given 5/6 nephrectomy and high phosphorus diet to make model.The model rats were divided into model group,Zuogui pill group.In addition,we set up blank control group at the same time.After operation,the blank control group was given normal drinking water,and the model group and Zuogui pill group were given high phosphorus drinking water.4 weeks after surgery,the blank control group and the model group were given sterile water for injection by gastric perfusion,and the Zuogui pill group was given Zuogui pill aqueous solution by gastric perfusion for 8 weeks.Rats were sacrificed after 8 weeks of administration.Serum BUN,Scr and IL-1βwere detected.The expressions of IL-1βand p65 in thoracic aorta of rats were detected by immunohistochemistry.Von kossa staining was used to observe the calcification of thoracic aorta in rats.Results:After 8 weeks of administration,BUN in the blank control group and Zuogui pill group was statistically significant compared with the model group(P<0.05).Compared with the model group,Scr in the blank control group and Zuogui pill group was statistically significant(P<0.01,0.05 respectively).IL-1βin the blank control group and Zuogui pill group was statistically significant compared with that in the model group(P<0.01).There was almost no expression of IL-1βand p65 in the thoracic aorta of rats in the blank control group.The expression of IL-1βand p65 in the model group was significantly higher than the Zuogui pill group,but significantly lower than the model group.Von kossa staining showed that there was no obvious calcium salt deposition in the intima of the thoracic aorta in the blank control group.The calcium salt deposition in the aorta of the model group was obvious,and the calcium salt deposition in the Zuogui pill group was significantly reduced compared with the model group.Conclusion:Zuogui pill can improve the calcification of
