摘  要:生长素响应因子(auxin response factor,ARF)是真核生物中普遍存在的转录因子之一,主要调控生长素应答基因的表达,与植物的生长发育和信号转导等多个生理过程密切相关。从桑树基因组数据库中共鉴定了16个ARF基因家族成员。理化性质分析表明桑树ARF蛋白的氨基酸个数为606~1144,分子质量为67.35~127.75 kD,等电点为4.86~8.37,大部分表现为酸性且都为亲水蛋白。根据系统发育及蛋白质结构分析,可将桑树ARF成员分为4个亚家族,同一亚族中的成员具有相同保守基序及类似基因结构,ARF蛋白至少具备B3、Auxin_resp、AUX_IAA等结构域中的一种;表达模式分析表明,ARF基因在桑树的根、桑皮、叶、芽、花等组织中的时空表达存在显著差异,在芽中呈现较高的表达,这可能与桑树芽器官的细胞分化程度大密切相关;MnARF9、MnARF10、MnARF11、MnARF12、MnARF15和MnARF16等6个基因在桑芽不同发育阶段的转录水平呈逐渐下调趋势,表明这些基因可能在桑芽前期的生长发育和细胞分化等生理反应中发挥作用。上述结果为后续桑树ARF家族基因的功能和进化分析以及桑树分子育种及良种选育等研究工作提供了一定的理论基础。Auxin response factor(ARF)is one of the ubiquitous transcription factors in eukaryotes.It mainly regulates the expression of auxin genes and is closely related to many physiological processes such as plant growth and development and signal transduction.A total of 16 ARF gene family members were identified from mulberry genome database.Physicochemical analysis showed that the amino acid number of the encoded protein ranged from 606 to 1144,the molecular mass was 67.35-127.75 kD,and the isoelectric point was 4.86-8.37.Most of ARF proteins were acidic and hydrophilic.According to phylogenetic and protein structure analysis,ARF members of mulberry could be divided into 4 subfamilies,members of the same subfamily had the same conserved motif and similar gene structure,and ARF proteins had at least one of the structural domains such as B3,Auxin_resp and AUX_IAA.Expression pattern analysis showed that there were significant differences in the spatiotemporal expression of ARF genes in the root,epidermis,leaf,bud,flower and other tissues of mulberry,with a relative higher expression level in the bud,which may be closedly related to the high degree of cell differentiation in the bud of mulberry.The transcription levels of 6 genes,including MnARF9,MnARF10,MnARF11,MnARF12,MnARF15 and MnARF16,showed a downward trend in different developmental stages of mulberry bud,indicating that these genes may play a role in the physiological reaction of growth and cell differentiation in the early stage of mulberry bud.The above results provided a theoretical basis for further research on the function and evolution of ARF family genes in mulberry,molecular breeding and improved variety breeding of mulberry.
