摘 要:多糖是药用真菌桑黄的主要活性成分之一。以2,4,6-三硝基苯磺酸(TNBS)诱导大鼠产生溃疡性结肠炎(UC),造模翌日连续9 d灌胃给予桑黄多糖进行干预,初步揭示桑黄多糖对溃疡性结肠炎的治疗效果及作用机制。结果显示,与模型大鼠比较,桑黄多糖各剂量组的大鼠体质量下降、粪便性状指数及结肠组织病变程度均显著减轻,结肠长度显著增加(P<0.01),单位长度质量显著降低(P<0.01),血清中的白介素6(IL-6)、肿瘤坏死因子α(TNF-α)和丙二醛(MDA)含量均显著下降(P<0.05),白介素10(IL-10)含量显著增加(P<0.05);结肠NF-κB p65蛋白表达水平以及IL-6和TNF-α mRNA表达水平均显著降低(P<0.05),IL-10 mRNA表达水平显著升高(P<0.05),剂量关系明确。结果表明,桑黄多糖对大鼠实验性溃疡性结肠炎具有明显的治疗效果,作用机制可能与调节炎性细胞因子分泌、抗氧化和抑制NF-κB的表达有关。Polysaccharides are the main active ingredients of medicinal fungus Phellinus igniarius.In order to preliminarily reveal the therapeutic effect and mechanism of Phellinus igniarius polysaccharide on ulcerative colitis,ulcerative colitis(UC)in rats was induced by 2,4,6-trinitrobenzenesulfonic acid(TNBS),and the rats were given intragastric administration of Phellinus igniarius polysaccharide(PIP)for 9 consecutive days post TNBS administration.The experimental results showed that compared with the model rats,Phellinus igniarius polysaccharide in each dose group alleviated the body weight loss,fecal index and colon tissue lesions of the rats with ulcerative colitis,significantly increased the colon length(P<0.01),significantly decreased the mass per unit length of colon(P<0.01),significantly reduced the contents of interleukin-6(IL-6),tumor necrosis factor TNF-αand malondialdehyde(MDA)(P<0.05)and significantly increased interleukin-10(IL-10)in serum(P<0.05).The expression level of NF-κB p65 protein and IL-6,TNF-αmRNA in colonic mucosa were significantly decreased(P<0.05),while that of IL-10 mRNA was significantly increased with a clear dose relationship(P<0.05).All the above results showed that PIP had obvious therapeutic effect on experimental ulcerative colitis in rats,and its mechanism may be related to regulating the secretion of inflammatory cytokines,antioxidation and inhibiting the expression of NF-κB.
