摘  要:目的利用同步辐射显微断层成像(SRμCT)表征大鼠脊髓挫伤(SCC)后髓内微血管在慢性恢复期的三维形态计量学特点。方法成年雄性SD大鼠(取自中南大学动物实验中心)48只, 按随机数字表法将其分为对照组、SCC后12周和24周组(n=16/组)。SCC后12周和24周组采用改良的Allen’s打击法建立SCC模型, 对照组仅行椎板切除术。3组大鼠在相应时间点行脊髓血管灌注造影并取标本行SRμCT扫描成像(n=8/组)和血管免疫荧光染色(n=8/组), 组间均值比较采用单因素方差分析。结果在SCC后的慢性恢复期, 大鼠后肢BMS评分仅恢复至正常水平的61.1%~64.7%, 髓内血管再生进入平台期。RECA-1+血管密度在SCC后24周和12周组比较, 差异无统计学意义[(113.12±21.30)/mm2比(100.25±21.81)/mm2, t=1.194, P>0.05], 血管容积比在SCC后24周和12周组比较, 差异无统计学意义[(14.35±1.72)%比(12.61±1.87)%, t=1.936, P>0.05], 分支节点密度在SCC后24周和12周组比较, 差异无统计学意义[(4 600.0±1 319.1)/mm^(3)比(4 025.0±1 513.5)/mm^(3), t=0.810, P>0.05), 中央沟动脉血管分支计数在SCC后24周和12周组比较, 差异无统计学意义[(120.0±29.8)/mm^(3)比(126.3±32.1)/mm^(3), t=-0.354, P>0.05]。结论大鼠SCC后慢性期后肢运动功能的恢复进入平台期可能与脊髓髓内微血管内源性再生的终止有关。Objective To characterize the three-dimensional morphometric characteristics of spinal microvascular network by using the synchrotron radiation-based micro-computed tomography(SRμCT)in the chronic recovery period after spinal cord contusion(SCC)in rats.Methods Totally,48 adult male Sprague-Dawley(SD)rats(from Animal Experimental Center of Central South University)were randomly divided into control group,12-week and 24-week groups after SCC by a table of random number(n=16/group).The 12-week and 24-week groups after SCC were established by improved Allen's method,and the control group only underwent laminectomy.The rats in three groups underwent SCC angiography and the samples were taken for SRμCT scanning(n=8/group),and immunofluorescence staining(n=8/group).The mean values of the groups were compared by a one-way analysis of variance(ANOVA).Results During the chronic recovery after SCC,the hind limb Basso Mouse Scale(BMS)only recovered to 61.1%-64.7% of the normal level,and the angiogenesis entered a plateau stage.Rat endothelial cell antigen-1+(RECA-1+)vessel density did not differ significantly between 24 and 12 groups after SCC[(113.12±21.30)/mm^(2)vs.(100.25±21.81)/mm^(2),t=1.194,P>0.05],vessel volume ratio did not differ significantly between 24 and 12 groups after SCC[(14.35±1.72)%vs.(12.61±1.87)%,t=1.936,P>0.05],and segmental node density did not differ significantly between 24 and 12 groups after SCC[(4600.0±1319.1)/mm^(3)vs.(4025.0±1513.5)/mm^(3),t=0.810,P>0.05].There was also no significant difference between the 24 and 12 weeks contusion group in central sulcus artery(CSA)segmental count[(120.0±29.8)/mm^(3)vs.(126.3±32.1)/mm^(3),t=-0.354,P>0.05].Conclusion In chronic recovery stage after SCC,the recovery of motor function of hind limb of rats entered a plateau stage,which may be related to the termination of angiogenesis of intramedullary microvascular network in spinal cord.
