摘  要:目的:通过高效液相色谱法同时测定小秦艽花主要药效成分的含量并联合多元统计分析,建立小秦艽花的质量评价方法。方法:采用高效液相色谱法测定15批小秦艽花药材中6种成分的含量,并对结果进行聚类分析及主成分分析。采用依利特hypersil ODS C_(18)分析用色谱柱,乙腈(B)-水(A)为流动相进行梯度洗脱;检测波长:獐牙菜苦苷、马钱苷为240 nm,龙胆苦苷、异荭草苷、异牡荆苷、齐墩果酸为210 nm;柱温为30℃。结果:獐牙菜苦苷、马钱苷分别在94.8~948μg(r=0.9999)、74.64~746.4μg(r=0.9999)浓度范围内与峰面积呈良好的线性关系。龙胆苦苷、异荭草苷、异牡荆苷、齐墩果酸分别在77.52~775.2μg(r=0.9999)、76.56~765.6μg(r=0.9999)、101.28~1 012.8μg(r=0.9999)、32.4~324μg(r=0.9999)浓度范围内与峰面积呈良好的线性关系。獐牙菜苦苷、马钱苷平均回收率分别为100.01%、99.91%,龙胆苦苷、异荭草苷、异牡荆苷、齐墩果酸平均回收率分别为100.18%、100.28%、98.40%和97.32%。小秦艽花药材中獐牙菜苦苷、马钱苷平均含量分别为4.85、1.02 mg/g。龙胆苦苷、异荭草苷、异牡荆苷、齐墩果酸平均含量分别为2.33、7.43、3.22、0.96 mg/g。聚类分析法将15批小秦艽花药材分为两大类,不同地区小秦艽花中6种成分的含量差异明显。SPSS 22.0软件主成分分析结果显示,主成分1(龙胆苦苷)及主成分2(异荭草苷)的整体方差贡献率达到了86.347%;R语言统计分析软件主成分分析结果显示,主成分1及主成分2的方差贡献率累积方差贡献率为95.2%,这与SPSS软件分析结果基本一致。结论:本测定方法快速、简便,重复性好,可为小秦艽花药材的质量控制提供依据。Objective:The quality evaluation method of Gentiana macrophylla was established by simultaneous determination of the main pharmacodynamic components of Gentiana macrophylla by HPLC and multivariate statistical analysis.Methods:The contents of 6 components in 15 batches of Gentiana macrophylla were determined by HPLC,and the results were analyzed by cluster analysis and principal component analysis.Elite hypersil ODS Cis was used as the chromatographic column for analysis,acetonitrile(B)-water(A)was used as the mobile phase for gradient elution;Detection wavelength:swertiamarin and loganin were 240 nm,gentiopicroside,isocarcinoside,isovitexin and oleanolic acid were 210 nm;The column temperature is 30 C.Results:Swertiamarin and loganin had a good linear relationship with the peak area,witches at 94.8-948μg(r=0.9999),74.64-746.4μg(r=0.9999)concentration range.Gentiopicroside,isomangoside,isovitexin and oleanolic acid had a good linear relationship with the peak area,witches at 77.52-775.2μg(r=0.9999),76.56-765.6μg(r=0.9999),101.28-1012.8μg(r=0.9999),32.4-324μg(r=0.9999)concentration range.The average recoveries of swertiamarin and loganin were 100.01%,99.91%respectively,the average recoveries of gentiopicroside,isomangoside,isovitexin and oleanolic acid were 100.18%,100.28%,98.40%,97.32%respectively.The average contents of swertiamarin and loganin in Gentiana macrophylla were 4.85,1.02 mg/g.The average contents of gentiopicroside,isomangoside,isovitexin and oleanolic acid were 2.33,7.43,3.22,0.96 mg/g.The results of cluster analysis showed that 15 batches of Gentiana macrophylla could be clustered into two categories,there were significant differences in the contents of six components in the flowers of Gentiana macrophylla in different regions.The principal component analysis results of SPSS 22.0 software show that the overall variance contribution rates of principal components 1(gentiopicroside)and 2(isomangoside)reached 86.347%;The principal component analysis results of R language statistical software show
