摘 要:目的:初步探讨败酱草对小鼠急性酒精性肝损伤的作用。方法:小鼠随机分为空白组、模型组、硫普罗宁组、败酱草高剂量组、败酱草中剂量组、败酱草低剂量组。预防性给药:败酱草高剂量组、败酱草中剂量组、败酱草低剂量组每日分别灌胃败酱草;硫普罗宁组每日分别灌胃硫普罗宁;空白组和模型组每日分别灌胃等体积蒸馏水。1周后,造模:模型组、硫普罗宁组、败酱草高剂量组、败酱草中剂量组、败酱草低剂量组50%乙醇溶液灌胃,空白组等体积蒸馏水灌胃。苏木素-伊红染色后观察肝脏组织切片病变程度;检测血清丙氨酸氨基转移酶(Alanine Aminotransferase,ALT)、天门冬氨酸氨基转移酶(Aspartate Aminotransferase,AST)、超氧化物歧化酶(Superoxide Dismutase,SOD)、丙二醛(Malonic Dialdehyde,MDA)、谷胱甘肽过氧化物酶(Glutathione Peroxidase,GSH-Px)、肿瘤坏死因子-α(Tumor Necrosis Factor-α,TNF-α)、白细胞介素(Interleukin,IL)-1β、IL-6水平。结果:与空白组相比,模型组ALT、AST水平明显升高(P<0.05);MDA水平明显升高(P<0.05);TNF-α、IL-6、IL-1β水平明显升高(P<0.05);SOD、GSH-Px水平明显降低(P<0.05)。与模型组相比,败酱草高剂量组、败酱草中剂量组、败酱草低剂量组和硫普罗宁组ALT、AST水平明显降低(P<0.05);MDA水平明显降低(P<0.05);TNF-α、IL-1β、IL-6水平明显降低(P<0.05);SOD、GSH-Px水平明显升高(P<0.05)。结论:败酱草对于急性酒精性小鼠肝损伤有保护作用。bjective:To study the effect of patrinia on acute alcoholic liver injury in mice.Methods:The selected mice were randomly divided into the blank group,the model group,the tiopronin tablet group,the high-dose patrinia group,the medium-dose patrinia group,and the low-dose patrinia group.The mice in the patrinia groups were gavaged daily.The mice in the tiopronin tablet group was given tiopronin tablet gavage daily.The mice in the blank group and the model group were gavaged with equal volume of distilled water for one week.The mice in the model group,the tiopronin tablet group,and the patrinia groups were given 50%ethanol solution by gavage,and the blank group was given equal volume of distilled water.After hematoxylin-eosin staining,the pathological degree of liver tissue lesion section was observed.The levels of ALT,AST,SOD,MDA,GSH-Px,TNF-α,IL-1βand IL-6 were detected.Results:Compared with the blank group,the levels of ALT and AST in the model group were significantly increased(P<0.05),the MDA level was significantly increased(P<0.05),the TNF-α,IL-6 and IL-1βlevels were significantly increased(P<0.05),the SOD and GSH-Px levels were decreased significantly(P<0.05).Compared with the model group,the levels of ALT and AST in the patrinia groups and the tiopronin tablet group were significantly decreased(P<0.05),the MDA level was decreased significantly(P<0.05),the TNF-α,IL-6 and IL-1βlevels were significantly decreased(P<0.05),the SOD and GSH-Px levels were increased significantly(P<0.05).Conclusion:Patrinia shows a certain protective effect on mice with alcoholic liver injury.
