摘 要:目的利用白及多糖的疏水改性技术,将其自组装成具有良好性能的纳米粒。方法采用透析法制备胆甾醇琥珀酰基白及多糖自组装纳米粒。通过稳态荧光探针法、透射电镜、动态光散射、X射线衍射以及差示扫描量热法对白及多糖及其衍生物进行物理表征。结果经稳态荧光探针法测定不同取代度胆甾醇琥珀酰基白及多糖-临界胶束浓度分别为0.0794、0.0398、0.0316 mg·ml^(-1),结果表明取代度越大临界胶束质量浓度越小;对透射电镜、动态光散射纳米粒子的形态特征及粒径进行了表征,表明取代度越大,纳米粒子粒径越小。通过X射线衍射和差示扫描量热法的物理表征,结果表明胆甾醇琥珀酸酯是小分子晶体,白及多糖、胆甾醇琥珀酰基白及多糖及其纳米粒结晶性较差。结论本方法成功制备了胆甾醇琥珀酰基白及多糖自组装纳米粒,制备方法简单可靠,为后续进一步研究奠定基础。Objective To prepare self-assembled nanoparticles from Bletilla Striata polysaccharide with favorable physiochemical properties via hydrophobic modification.Methods The cholesterol succinyl Bletilla Striata polysaccharide self-assembled nanoparticles were prepared through dialysis.The product as well as its derivatives was characterized using the steady-state fluorescence probe method,transmission electron microscope imaging,dynamic light scattering,X-ray diffraction and differential scanning calorimetry.Results The critical micelle concentrations of cholesterol succinyl Bletilla Striata polysaccharide with different degrees of substitution were determined as 0.0794,0.0398 and 0.0316 mg·ml^(-1) by the steady-state fluorescence probe method,which demonstrated a negative correlation between critical micelle concentrations and degrees of substitution.The results from transmission electron microscope imaging and dynamic light scattering indicated negative relationships between degrees of substitution and particle size.Although the cholesteryl succinate appeared to be small molecule crystals,the crystallinity of Bletilla Striata polysaccharide and its cholesterol succinyl derivatives was poor before and after formatting nanoparticles according to the X-ray diffraction and differential scanning calorimetry readout.Conclusion The cholesterol succinyl Bletilla Striata polysaccharide self-assembled nanoparticles are prepared in a simple and reliable preparation process.
