摘  要:目的:研究和完善根状生长实验方法,降低蜡样芽孢杆菌检验结果假阳性率。方法:根据GB 4789.14—2014,在单因素变量情况下,筛选根状生长实验最佳条件。选择蜡样芽胞杆菌标准菌株、蕈状芽胞杆菌标准菌株、实验室分离株,用普通营养琼脂培养基(Nutrient Agar,NA)、平板计数琼脂培养基(Plate Count Agar,PCA)、胰酪大豆胨琼脂培养基(Tryptose Soya Agar,TSA)、含0.6%酵母膏的胰酪胨大豆琼脂培养基(Trypticase Soy-Yeast Extract Agar,TSA-YE),在不同条件下分别进行根状生长实验。结果:经30℃培养72 h,PCA倾注9 mL的琼脂量,蜡样芽胞杆菌与蕈状芽胞杆菌根状生长实验效果最好,不易出现假阳性。结论:单一变量情况下,PCA>NA>TSA>TSA-YE,倾注的培养基厚度为9 mL>12 mL>15 mL>18 mL>20 mL,30℃培养效果好于37℃,培养时间为72 h。该实验条件优化为区分蜡样芽胞杆菌和蕈状芽胞杆菌提供了一定的理论依据。Objective:Research and improve the experimental method of root growth to reduce the false positive rate of Bacillus cereus test results.Method:According to GB 4789.14—2014,under the condition of single factor variables,the optimal conditions for rhizoid growth experiment were screened.Standard strains of Bacillus cereus,standard strains of Bacillus fungoides and laboratory isolates were selected to conduct root growth experiments under different conditions with common nutrient nutrient agar medium(NA),plate counting agar medium(PCA),tryptose soya agar medium(TSA)and trypsin soy-yeast extract agar medium(TSA-YE)containing 0.6%yeast extract.Result:After culture at 30℃for 72 h,9 mL agar was poured into PCA,the results of Bacillus cereus and Bacillus fungoides root growth experiment were the best,and false positivity was not easy to appear.Conclusion:Under single variable conditions,PCA>NA>TSA>TSA-YE,and the thickness of poured medium was 9 mL>12 mL>15 mL>18 mL>20 mL.The culture effect at 30℃was better than that at 37℃,and the culture time was 72 h.The optimization of the experimental conditions provides a theoretical basis to distinguish the wax form from the mushroom form.
