摘 要:[目的]探讨线粒体COI基因对叶尔羌河鱼类资源鉴定及遗传多样性分析的适应性。[方法]分析叶尔羌河5种鲤科的50条COI基因片段序列,并结合GenBank比对筛选出的12条高同源序列基于邻接法构建系统进化树。[结果]5种鱼类COI基因同源序列的长度675 bp,碱基组成展现出显著A/T的偏倚性,转换/颠换的比值(R值)为3.2;核苷酸变异位点有146个,定义了10个单倍型;除塔里木裂腹鱼和宽口裂腹鱼物种间的遗传距离(0.002 4),其余物种间的遗传距离均符合2%阈值或10倍原则;系统进化树中,除塔里木裂腹鱼和宽口裂腹鱼外,斑重唇鱼、鲫鱼和鲤鱼均形成独立分支,推测两者亲缘关系较近或物种间未达到分化程度。[结论]COI基因适用于叶尔羌河除裂腹鱼属外鱼类的物种鉴定及多样性分析。[Objective]To explore the adaptability of mitochondrial COI gene to fish resource identification and genetic diversity analysis in Yarkant River.[Method] 50 COI gene fragment sequences of 5 species of Cyprinidae in Yarkant River were analyzed,and the phylogenetic tree was constructed based on the neighbor-joining method combined with 12 high homologous sequences screened by GenBank alignment.[Result]The length of COI gene homologous sequence of 5 fish species was 675 bp,showing significant base bias,and the conversion/transmutation ratio(R value) was 3.2.There were 146 nucleotide mutation sites and 10 haplotypes were defined.Except for the genetic distance between Tarim schizostomus and wide-mouth schizostomus species(0.002 4),the genetic distance between other species conforms to the 2% threshold or 10-fold principle.In the phylogenetic tree,except for schizostomus Tarim and schizostomus broadmouth,all of them formed independent branches,suggesting that the two species were closely related or did not reach the degree of differentiation.[Conclusion]COI gene was suitable for species identification and diversity analysis of fishes except schizothorax in the Yarkant River.
