摘  要:目的探讨miR-125b-5p通过靶向BACH1增强β-地中海贫血(β-thal)单核细胞吞噬活性的机制。方法收集健康儿童与HbE型β-thal(HbE/β-thal)患儿外周血,流式细胞仪分析外周血单核细胞(PBMC)表型,qRT-PCR法测定miR-125b-5p的表达量,分析HbE/β-thal患儿miR-125b-5p表达与病情及激活表型的关系,双荧光素酶报告基因系统检验miR-125b-5p与BACH1的靶向关系,western blot检测BACH1与HO-1的表达量,在正常PBMC中同时上调miR-125b-5p与BACH1,在HbE/β-thal来源PBMC中同时下调miR-125b-5p与BACH1,测定PBMC吞噬活性的变化。结果2组儿童PBMC数量没有明显差异(P>0.05),但HbE/β-tha患儿CD14hiCD16+表型PBMC比例增加(P<0.05),HbE/β-thal患儿PBMC内miR-125b-5p表达量显著高于健康儿童(P<0.05),miR-125b-5p的表达与RBC计数、Hb含量、Hct、MCH均呈负相关(P<0.05),而与CD14hiCD16+比例呈现正相关(P<0.05);双荧光素酶报告基因系统与western blot证实miR-125b-5p能靶向抑制BACH1表达(P<0.05),且与正常儿童比较,HbE/β-thal患儿PBMC中BACH1表达量降低(P<0.05),而HO-1表达量升高(P<0.05);miR-125b-5p上调可以显著增强健康儿童来源PBMC的吞噬能力(P<0.05),而过表达BACH1能逆转该情况(P<0.05),相反,抑制miR-125b-5p能降低具有高吞噬能力的HbE/β-thal患儿来源PBMC的吞噬活性(P<0.05),而抑制BACH1能逆转该现象(P<0.05)。结论miR-125b-5p通过抑制BACH1来增强HbE/β-thal患儿PBMC吞噬活性。Objective To investigate the role of miR-125b-5p in enhancing the phagocytic activity of monocytes in children withβ-thalassemia by targeting BACH1.Methods Peripheral blood samples of healthy children and children with hemoglobin E/β-thalassemia(HbE/β-thal)were collected foRidentifying the phenotype of peripheral blood monocytes(PBMCs)via flow cytometry.The expression of miR-125b-5p was measured by quantitative reverse transcription polymerase chain reaction(qRT-PCR).The correlation of miR-125b-5p expression with disease condition and activation phenotype in children with HbE/β-thal was identified.The targeting relationship between miR-125b-5p and BACH1 was tested by dual-luciferase reporteRassay.Protein expressions of BACH1 and HO-1 were detected by Western blot.Changes of phagocytic activity of PBMCs isolated from healthy children and transfected with overexpression plasmids of both miR-125b-5p and BACH1,and those isolated from children with HbE/β-thal and transfected with knockdown plasmids of both miR-125b-5p and BACH1 were measured.Results There was no significant difference in the numbeRof PBMCs between the two groups(P>0.05),but the proportion of PBMCs with CD14hiCD16+phenotype was significantly higheRin children with HbE/β-thal(P<0.05).The expression of miR-125b-5p in PBMCs of children with HbE/β-thal was significantly higheRthan that of healthy children(P<0.05).The expression of miR-125b-5p was negatively correlated with the red blood cell(RBC)count,hemoglobin(Hb)level,hematocrit(Hct)and mean corpusculaRhemoglobin(MCH)in children with HbE/β-thal,but positively correlated with the proportion of CD14hiCD16+(P<0.05).Dual-luciferase reporteRassay and Western blot confirmed that miR-125b-5p could target BACH1 and inhibit its expression(P<0.05).Compared with those in healthy children,BACH1 was downregulated and HO-1 was upregulated in PBMCs of children with HbE/β-thal(P<0.05).Overexpression of miR-125b-5p significantly enhanced the phagocytosis of PBMCs isolated from healthy children,which could be re
