摘  要:为制备不同含量沙枣提取物(EA)的静电纺丝纳米纤维敷料,利用静电纺丝法制备了聚偏氟乙烯/沙枣提取物(PVDF/EA)纳米纤维膜。通过SEM、FTIR分析其微观形貌和化学结构。研究了纳米纤维膜的力学性能、体外药物释放性、抗氧化性以及抗菌性,并对PVDF/EA0.4纳米纤维膜的抑菌机理进行了初步分析。研究结果表明:所制备PVDF/EA纳米纤维膜的纤维直径呈现先增后减的趋势,并在EA质量浓度为0.6 mg/m L时直径达到最大,为(846±181)nm;纳米纤维膜在干、湿状态下的拉伸强度介于(5.25±0.33)~(9.71±0.20)MPa,可满足伤口敷料的力学要求。纳米纤维膜中EA累计释放量随负载质量浓度的增加而增大,抗氧化性与EA含量成正比,最大可达76.92%。EA对大肠埃希菌、金黄色葡萄球菌的抑制率均在99%以上。通过对细菌的荧光染色发现,EA能够通过破坏细胞膜杀菌。所制备的PVDF/EA纳米纤维膜具备作为伤口敷料的潜力。To prepare electrospun nanofibrous dressings containing different contents of Elaeagnus angustifolia extract(EA),PVDF/EA nanofibrous membranes are prepared by the electrospinning method.The microscopic morphology and chemical structure are analyzed by SEM and FTIR.The mechanical properties,in vitro drug release,antioxidant and antibacterial properties of nanofiber membranes are studied,and the antibacterial mechanism of PVDF/EAO.4 nanofiber membranes is preliminarily analyzed.The results show that the diameter of the prepared PVDF/EA nanofiber membrane shows a trend of increasing first and then decreasing,and the diameter is the largest at 846+181 nm when the loading concentration of EA is 0.6 mg/mL.The tensile strength is between 5.25+0.33 and 9.71±0.20 MPa,which meets the mechanical requirements for wound dressings.In addition,the cumulative release of EA in the nanofiber membrane increases with increasing loading concentration,the antioxidant activity increases from 71.12%to 76.92%,and the inhibition rates against Escherichia coli and Staphylococcus aureus are both above 99%.Through fluorescent staining of bacteria,it is found that EA could destroy the cell membrane to kill bacteria and has good antibacterial properties.Therefore,the prepared PVDF/EA nanofiber membrane has potential as a wound dressing.
