摘  要:目的探讨circ_0101504/miR-454-3p/ARAP2通路在胃癌生长中的作用及其机制。方法选取2020年5月至2020年12月入上海交通大学医学院附属瑞金医院卢湾分院行胃癌切除手术的癌组织及癌旁组织标本30对,采用实时荧光定量PCR检测组织和细胞中circ_0101504和miR-454-3p的表达,MTT检测胃癌细胞BGC-823增殖,Transwell检测胃癌细胞BGC-823迁移,流式细胞仪检测胃癌细胞BGC-823凋亡和细胞周期,荧光素酶报告基因检测circ_0101504和miR-454-3p及miR-454-3p和ARAP2之间的相互作用。结果胃癌组织中circ_0101504的水平明显高于癌旁组织;胃癌组织中miR-454-3p的水平明显低于癌旁组织;与GES-1相比,胃癌细胞系MKN-28、SNU-1、HGC-27、BGC-823中circ_0101504表达明显升高,胃癌细胞系MKN-28、SNU-1、HGC-27、BGC-823中miR-454-3p表达明显降低;与si-NC组相比,si-circRNA_0101504组中的circRNA_0101504表达明显降低,miR-454-3p表达显著升高;与si-NC组相比,si-circRNA_0101504组细胞增殖能力、迁移数量显著降低,细胞凋亡率、G0/G1期细胞比例及ARAP2蛋白表达显著增加,S期细胞比例显著降低;与si-circRNA_0101504+miR-NC组相比,si-circRNA_0101504+miR-454-3p抑制剂组的细胞增殖能力、迁移数量明显升高,细胞凋亡率、G0/G1期细胞比例及ARAP2蛋白表达明显降低,S期细胞比例明显上升,与miR-NC组相比,miR-454-3pmimics组中的相对荧光素酶活性显著降低,miR-454-3p mimic减少ARAP2-wt荧光素酶活性。结论circ_0101504/miR-454-3p/ARAP2通路在胃癌的发生发展中起着至关重要的作用,可能成为胃癌诊断及治疗的新靶点。Objective To investigate the role and mechanism of circ_0101504/miR-454-3p/ARAP2 pathway in the growth of gastric cancer.Methods Thirty pairs of cancer tissue and adjacent tissue samples were selected from our hospital for gastric cancer resection from May 2020 to December 2020.Real-time quantitative PCR was used to detect the expression of circ_0101504 and miR-454-3p in tissues and cells,and MTT was used to detect the proliferation of GC-823 cells.Transwell was used to detect BGC-823 migration,flow cytometry was used to detect apoptosis and cell cycle of BGC-823,luciferase reporter gene was used to detect circ_0101504 and miR-454-3p,and interaction between miR-454-3p and ARAP2.Results The level of circ_0101504 in gastric cancer tissue was significantly higher than that in paracancer tissue.The level of miR-454-3p in gastric cancer tissues was significantly lower than that in paracancer tissues.Compared with GES-1,the expression of circ_0101504 in gastric cancer cell lines MSN-28,SNU-1,HGC-27 and BGC-823 was significantly increased,while the expression of miR-454-3p in gastric cancer cell lines MSN-28,SNU-1,HGC-27 and BGC-823 was significantly decreased.Compared with the si-NC group,the expression of circRNA_0101504 in si-circRNA_0101504 group was significantly decreased,while the expression of miR-454-3p was significantly increased.Compared with the si-NC group,the cell proliferation ability and migration number of si-circRNA_0101504 group were significantly decreased,the cell apoptosis rate,the proportion of G0/G1 phase cells and the expression of ARAP2 protein were significantly increased,and the proportion of S phase cells were significantly decreased.Compared with si-circRNA_0101504+miR-NC group,si-circRNA_0101504+miR-454-3p inhibitor group significantly increased cell proliferation and migration,and significantly decreased cell apoptosis rate,G0/G1 phase cell proportion and ARAP2 protein expression.Compared with the miR-NC group,the relative luciferase activity of the miR-454-3pmimics group decreased signif
