摘 要:为筛选一套便捷高效的‘黎平2号’油茶良种组培快繁技术体系以应用于生产,本研究以贵州小果油茶良种‘黎平2号’带芽茎段为试验材料,以MS为基本培养基,加入不同组合的植物激素进行‘黎平2号’油茶组培快繁技术研究。结果表明:MS+2.5 mg·L^(-1)6-BA+0.8 mg·L^(-1)NAA+0.08 mg·L^(-1)TDZ为‘黎平2号’不定芽诱导最佳培养基,MS+0.03 mg·L^(-1)NAA+0.02 mg·L^(-1)TDZ为最佳增殖培养基,1/2MS+0.5 mg·L^(-1)IBA+0.5 mg·L^(-1)NAA为‘黎平2号’油茶的最佳生根培养基,本研究结果将为贵州省油茶良种产业化发展提供种苗保障。An efficient and convenient system is highly required for the tissue culture and rapid propagation in recent years.This study aims to screen such a new system for the improved variety of'Liping No.2'Camellia oleifera in forest production.The experimental materials were taken as the sprouted stem segments of small fruit C.oleifera variety'Liping No.2'in Guizhou Province of China.Murashige and Skoog(MS)was then selected as the basic culture medium.Finally,an optimal combination of plant hormones was achieved for the better performance of the tissue culture and rapid propagation.The results showed that the MS+2.5 mg·L^(-1)6-BA+0.8 mg·L^(-1)NAA+0.08 mg·L^(-1)TDZ was the optimal medium for the inducing adventitious buds in'Liping No.2',while the MS+0.03 mg·L^(-1)NAA+0.02 mg·L^(-1)TDZ was the optimal proliferation medium,and 1/2MS+0.5 mg·L^(-1)IBA+0.5 mg·L^(-1)NAA was the optimal rooting medium for'Liping No.2'small fruit C.oleifera.The finding can provide a strong reference for the seed and seedling protection in the large-scale industrialization of C.oleifera in Guizhou Province of China.
