摘 要:目的探讨二苯乙烯苷(TSG)对血糖波动性糖尿病小鼠血清血管内皮生长因子(VEGF)、色素上皮衍生因子(PEDF)水平的影响及其与肾功能的关系。方法把30只小鼠随机分为正常组、血糖波动性糖尿病组、血糖波动性糖尿病TSG治疗组动物模型,每组10只小鼠。小鼠左下腹腔内注射链脲佐菌素(60mg/kg)复制糖尿病动物模型;糖尿病小鼠模型制备成功后,采用腹腔注射给予葡萄糖和胰岛素造成血糖波动;血糖波动性糖尿病TSG治疗组每天2次50 mg/kg TSG灌胃,正常组及血糖波动性糖尿病组给予等量的生理盐水灌胃,连续8周后,所有小鼠测空腹血糖;采用全自动生化分析仪测定血清血肌酐(Serum Creatinine,SCr)、血尿素氮(Blood urea nitrogen,BUN)含量;用ELISA法检测各组小鼠血清VEGF、PEDF的含量;用苏木精-伊红(HE)染色,光学显微镜观察各组小鼠肾组织的形态学变化。结果与正常组比较,血糖波动性糖尿病组、血糖波动性糖尿病TSG治疗组的空腹血糖[分别为(20.700±1.399)mmol/L、(19.220±1.132)mmol/L]、血清SCr[分别为(172.500±15.537)μmol/L、(151.600±12.912)μmol/L]、BUN[分别为(20.870±4.109)mmol/L、(13.760±2.000)mmol/L]、VEGF[分别为(80.330±5.081)pg/ml、(74.030±4.473)pg/ml]、PEDF[(39.640±4.617)pg/ml、(45.570±5.485)pg/ml]水平升高,差异有统计学意义(P<0.05);与血糖波动性糖尿病组比较,血糖波动性糖尿病TSG治疗组小鼠空腹血糖、血清SCr、BUN、VEGF水平降低,PEDF水平升高,差异有统计学意义(P<0.05)。HE染色结果显示:正常组小鼠肾组织结构正常,血糖波动性糖尿病组、血糖波动性糖尿病TSG治疗组的小鼠肾小球体积增大,肿胀,囊性扩张,间质水肿,有炎细胞浸润,肾小管出现萎缩、消失,血糖波动性糖尿病组病理变化更严重。结论TSG可改善血糖波动性糖尿病小鼠的血糖水平,增强肾脏对血肌酐、血尿素氮的清除,降低VEGF水平,升高PEDF水平,发挥对血糖波动�Objective To investigate the effect of 2,3,4,5’⁃tetrahydroxy⁃stilbene⁃2⁃0⁃β⁃D⁃glycoside(TSG)on serum VEGF and PEDF levels in diabetes mice with fluctuating blood glucose and its relationship with re⁃nal function.Methods Thirty mice were randomly divided into normal group,blood glucose fluctuation diabetes group,blood glucose fluctuation diabetes TSG treatment group,10 mice in each group.Streptozotocin(60 mg/kg)was injected into the left lower abdominal cavity of mice to replicate the animal model of diabetes.After the success⁃ful preparation of diabetes mouse model,glucose and insulin were injected intraperitoneally to cause blood glucose fluctuation.TSG 50 mg/kg was given twice a day by gavage in the TSG treatment group,and the normal group and the blood glucose fluctuation diabetes group were given the same amount of normal saline.After 8 consecutive weeks,fasting blood glucose was measured in all mice,serum creatinine(SCr)and blood urea nitrogen(BUN)were mea⁃sured by automatic biochemical analyzer.The serum VEGF and PEDF were detected by ELISA.HE staining and light microscope were used to observe the morphological changes of kidney tissue in each group.Results Compared with the normal group,the fasting blood glucose(20.700±1.399 mmol/L and 19.220±1.132 mmol/L,respectively),the levels of serum SCr(172.500±15.537μmol/L and 151.600±12.912μmol/L,respectively),BUN(20.870±4.109 mmol/L and 13.760±2.000 mmol/L,respectively),VEGF(80.330±5.081 pg/ml and 74.030±4.473 pg/ml,respective⁃ly)and PEDF(39.640±4.617 pg/ml and 45.570±5.485 pg/ml,respectively)in the blood glucose fluctuation diabetes group and the blood glucose fluctuation diabetes TSG treatment group were increased,and the difference was statisti⁃cally significant(P<0.05);Compared with the blood glucose fluctuation diabetes group,the fasting blood glucose,the levels of SCr,BUN and VEGF in serum of mice in the blood glucose fluctuation diabetes TSG treatment group were decreased,and the level of PEDF was increased,and the dif
