摘 要:水稻(Oryza sativa)中RIP基因家族编码典型的E3泛素连接酶,在生长发育、逆境响应过程中发挥重要作用。利用水稻基因组数据,采用生物信息学方法鉴定了水稻中的RIP基因家族成员,鉴定到6个RIP家族基因,命名为OsRIP1~6,其编码区长度在906~1086 bp之间,分布在5条染色体上,内含子数量为2~3个。水稻中RIP基因被分为3个亚家族,每个亚家族的内含子数量、基因结构、motif基本一致。蛋白结构域分析结果表明,在水稻中,RIP家族的所有成员均含有SINA和RING结构域。共线性分析显示,水稻与玉米存在最多的共线对,与拟南芥不存在共线对。对启动子顺式作用元件分析发现,RIP基因启动子区有多个非生物胁迫响应元件、光响应元件及激素响应元件。另外,还分析RIP基因家族在不同组织器官、不同发育时期及昼夜节律的表达模式,结果表明在根、茎及花器官中表达水平较高;OsRIP6在播种后83 d表达水平最高,而其他成员在播种后48~69 d的表达处于较高水平;昼夜表达的分析结果显示,在光照后10 h该基因家族成员的表达水平较高。在ABA、JA激素和干旱胁迫、盐胁迫处理下,OsRIP1~5表达水平上调,OsRIP6在JA、盐胁迫和甘露醇模拟干旱胁迫时表达被抑制。对水稻OsRIP基因家族进行了全面的分析,为进一步揭示OsRIP基因的生物学功能奠定基础。The RIP gene family in rice(Oryza sativa) encodes a typical E3 ubiquitin ligase that plays an important role in growth and development,and in response to stress.In this study,we used rice genomic data to identify the RIP gene family members in rice using bioinformatics methods,and identified six RIP family genes,named OsRIP1-6,with coding regions ranging from 906-1086 bp in length and distributed on five chromosomes with 2-3 introns.The RIP genes in rice were divided into three subfamilies,and the number of introns,gene structure,and motif of each subfamily were basically the same.The results of protein structural domain analysis showed that in rice,all members of this gene family contain SINA and RING structural domains.Synteny analysis showed that there were the most syntenic relationship with maize and no syntenic relationship with Arabidopsis.Analysis of promoter cis-acting elements revealed multiple abiotic stress response elements,light response elements and hormone response elements.This study also analyzed the expression patterns of the RIP gene family in different tissues and organs,different developmental periods and circadian rhythms,and the results showed that the expression levels were higher in root,stem and flower,OsRIP6 had the highest expression level at 83 days after sowing,while the highest expression of other members was at 48-69 days after sowing.The analysis of diurnal expression showed that the expression level of this gene family member was higher at 10 h after light.The expression levels of OsRIP1-5 were up-regulated under ABA,JA,drought stress and salt stress treatments,wheareas OsRIP6 expression was suppressed under JA,salt stress and mannitol treatment.This study provides a comprehensive analysis of the OsRIP gene family in rice and lays the foundation for further revealing the biological functions of OsRIP genes.
