摘 要:该试验旨在探讨珠美海棠试管苗室外瓶内生根的技术措施。4月中旬将接种了珠美海棠试管苗茎段的培养瓶分别放置在室外3种遮阴条件下培养,并于2016、2017、2018年连续3年的不同时期,在室外遮阴条件下进行多次珠美海棠试管苗瓶内生根培养试验。结果表明,在天津农学院所处气候条件下,4月中旬至5月下旬,在室外采用树荫、双层遮阳网或三层遮阳网遮阴等方式可以控制培养瓶内温度在35℃以下,培养瓶顶部光照在2000 lx以上,3种遮阴方式下珠美海棠试管苗的生根生长指标无显著差异,3种遮阴方式在该时期均可用于试管苗培养;室外采用树荫遮阴,4月下旬至7月初和9月初至10月中旬是珠美海棠试管苗瓶内生根的适宜时期,其中以4月下旬至6月上旬为最佳培养时期,生根率达到87.3%至97.8%。上述结果可为降低珠美海棠试管苗成本并促进其快繁技术的生产应用提供依据。The aim of this study was to investigate the in vitro rooting technique of Malus Zumi micro-shoots in outdoor environment.In middle ten days of April the culture bottles in which the micro-shoots were inoculated were respectively located under three shading conditions in outdoor environment for rooting culture,and many times experiments on rooting in vitro of Malus Zumi micro-shoots under shading in outdoor environment in 2016,2017 and 2018.The result showed that under climate condition in Tianjin agricultural university courtyard the temperature in culture bottle was controlled below 35℃and the light intensity on the bottle top was above 2000 lx using shading methods of tree shading,two course sunshade net shading or three course sunshade net shading from middle ten days of April to last ten days of May.There was no significant difference among the rooting and growth index of Malus Zumi plantlets under the three shading methods which could be all used for culturing the micro-shoots in this period.Using tree shading in outdoor the suitable time for rooting in vitro of Malus Zumi micro-shoots were the periods from last ten days of April to early July and from early September to middle ten days of October,and the most suitable period was from last ten days of April to first ten day of June,and in these periods the rooting rate of the micro-shoots reached 87.3%-97.8%.These results can be used as reference for reducing the cost of Malus Zumi plantlet in vitro and promoting applying the rapid technique of Malus Zumi to production.
