摘  要:探索奶牛乳腺炎粪肠球菌cylA基因对体外培养的单核巨噬细胞完全吞噬功能的影响。通过体外培养牛外周血单核巨噬细胞(BMC-7),利用牛乳腺炎粪肠球菌野生型菌株(BME1708)和cylA基因敲除突变株(BME1708ΔcylA)按照不同时间梯度侵染BMC-7,利用台酚蓝染色显微镜观察法,统计不同时间BMC-7对BME1708和BME1708ΔcylA的吞噬率和吞噬指数的差异。结果表明随着时间的延续,吞噬率和吞噬指数存在动态变化,至48 h时BMC-7对BME1708的吞噬率和吞噬指数分别为25%和2.3,而对BME1708ΔcylA的吞噬率和吞噬指数分别为50%和5.5。随着时间的进一步延续,巨噬细胞开始死亡。说明溶血素cylA基因阳性的粪肠球菌菌株对巨噬细胞的吞噬作用具有一定的负调控作用,并且在巨噬细胞内的存活率相对高,在单核巨噬细胞内存在一定的“内生”现象,这一现象可能是由该致病菌引起的牛乳腺炎常反复、难治愈的分子机制之一。To explore the effect of Enterococcus faecalis cylA gene on complete phagocytosis of monocyte macrophages cultured in vitro.By culturing bovine peripheral blood mononuclear macrophages(BMC-7)in vitro,BMC-7 was infected by Enterococcus faecalis wild-type strain(BME1708)and cylA gene knockout mutant(BME1708ΔcylA).The results showed that there was a dynamic change in the phagocytosis rate and phagocytosis index over time.At 48 h,the phagocytosis rate and phagocytosis index of BMC-7 to BME1708 were 25%and 2.3,respectively,while the phagocytosis rate and phagocytosis index of BME1708ΔcylA were 50%and 5.5,respectively.As time went on,macrophages began to die.This shows that Enterococcus faecalis with hemolysin cylA gene positive has a certain negative regulation effect on the phagocytosis of macrophages,and its survival rate in macrophages is relatively high.There is a certain"endogenous"phenomenon in monocyte macrophages,which may be one of the molecular mechanisms of bovine mastitis caused by this pathogen,which is often repeated and difficult to treat.
