摘 要:犬博卡病毒(CBoV)属于细小病毒科的无囊膜病毒,是一种从犬粪便中检测发现的新发病原,该病毒现已在多个国家和地区犬粪便中检测到,但其在上海地区的流行情况及其致病机制目前还不是很清楚。为了调查犬博卡病毒在上海犬中的流行情况,本研究于2019年6月至2019年12月,从上海市采集犬新鲜粪便206份,通过PCR扩增方法进行鉴定并进一步通过进化树对其基因组进行分析。结果显示在所搜集的206份样品中有3份为CBoV阳性(1.5%),为了对检测出的CBoV基因组特性进行分析,我们扩增出CBoV的全基因组,发现其基因组共含有5246个核苷酸,其中包含大小为249 bp的5'UTR、4853 bp的开放阅读框和144 bp的3'UTR。从系统进化树中可以看出该病毒与其他亚洲分离株亲缘关系较近,处于同一个分支,同属于CBoV2。本研究通过对CBoV的流行情况进行调查,揭示了CBoV在上海犬中的感染情况,为进一步研究CBoV的致病性及潜在的CBoV传染病的预警和防控提供科学依据和理论参考。Canine bocavirus(CBoV),a small non-enveloped virus,which was a newly described virus detected in the feces of dogs and classified as a genus within the Parvoviridae family.Canine Boka virus has been detected in dog feces in several countries and regions,but the prevalence and pathogenesis of the virus are poorly understood.To survey for CBoV infection in Shanghai,206 fecal samples from dogs were collected from June 2019 to December 2019.Using polymerase chain reaction(PCR),CBoVs were detected in 3 fecal samples(1.5%).To reveal the genetic characteristics of CBoVs,the full genome of CBoV was sequenced.The genome consists of 5246 nucleotides,excluding 249 bp of 5'UTR,4853 bp of open reading frame(ORF),and 144 bp(not containing poly A)of 3'UTR.Phylogenetic analysis showed that the CBoV was close to a previously circulated Asia isolate and clustered together with other CBoV2 strains.This study revealed the prevalence of CBoV in Shanghai dogs by investigating the infection of CBoV.The result also provided scientific evidence and theoretical reference for further study on the pathogenicity of CBoV and the early warning and prevention and control of potential CBoV infectious diseases.
