摘  要:目的 探讨miR-579-3p靶向角鲨烯单加氧酶(SQLE)对宫颈癌HeLa细胞铁死亡的影响,及其对宫颈癌HeLa细胞增殖、侵袭的作用机制。方法 将细胞分为H8组(正常培养H8细胞)、HeLa组(正常培养HeLa细胞)、HeLa+SQLE组(HeLa细胞过表达SQLE)和HeLa-SQLE+mimic-miR-579-3p组(过表达SQLE HeLa细胞转染mimic-miR-579-3p)。用细胞计数试剂盒-8(CCK-8)法检测细胞的增殖情况,用Transwell实验法检测细胞的迁移和侵袭情况,用实时荧光定量聚合酶链反应检测铁死亡相关基因的mRNA表达水平,用试剂盒法检测活性氧(ROS)和丙二醛(MDA)的水平。结果 H8组、HeLa组、HeLa+SQLE组和HeLa-SQLE+mimic-miR-579-3p组的第1天增殖率分别为35.71±1.87、54.63±3.49、95.86±2.92和46.98±3.51,侵袭细胞数分别为(24.14±2.54)、(101.00±6.63)、(146.71±12.02)和(42.71±10.40)个,迁移细胞数分别为(32.43±3.74)、(111.71±5.68)、(145.86±6.01)和(43.14±2.61)个,溶质载体家族7成员11 mRNA相对表达水平分别为1.00±0.07、11.22±1.74、25.59±2.06和2.43±0.45,谷胱甘肽过氧化物酶4 mRNA相对表达水平分别为1.00±0.11、8.83±0.33、16.03±3.17和1.97±0.09,ROS水平分别为(84.82±0.34)、(32.66±0.53)、(23.30±0.14)和(64.48±0.33)U·mL^(-1),MDA水平分别为(6.62±0.05)、(2.54±0.02)、(1.63±0.04)和(4.97±0.04)nmol·mL^(-1)。HeLa+SQLE组的上述指标与HeLa组和HeLa-SQLE+mimic-miR-579-3p比较,差异均有统计学意义(均P<0.05)。结论 miR-579-3p靶向SQLE可以促进宫颈癌HeLa细胞的铁死亡,从而抑制宫颈癌HeLa细胞的增殖和侵袭。Objective To investigate the effect of miR-579-3p targeting squalene epoxidase(SQLE)on ferroptosis of cervical cancer HeLa cells,and its mechanism of proliferation and invasion of cervical cancer HeLa cells.Methods The cells were divided into H8 group(H8 cells were cultured normally),HeLa group(HeLa cells were cultured normally),HeLa+SQLE group(HeLa cells with SQLE overexpression)and HeLa-SQLE+mimic-miR-579-3p group(HeLa cells with SQLE overexpression were transfected mimic-miR-579-3p).Cell proliferation was detected by cell counting kit-8(CCK-8)assay.Transwell was used to detect cell migration and invasion.The mRNA expressions of ferroptosis-related genes were detected by quantitative real-time polymerase chain reaction.The levels of reactive oxygen species(ROS)and malondialdehyde(MDA)were detected by kit method.Results In H8,HeLa,HeLa+SQLE and HeLa-SQLE+mimic-miR-579-3p groups,the proliferation rates on the first day were 35.71±1.87,54.63±3.49,95.86±2.92 and 46.98±3.51;the number of cells invaded were 24.14±2.54,101.00±6.63,146.71±12.02 and 42.71±10.4;the number of cells was32.43±3.74,111.71±5.68,145.86±6.01 and 43.14±2.61;the relative expression levels of solute carrier family7 member 11 mRNA were 1.00±0.07,11.22±1.74,25.59±2.06 and 2.43±0.45;the relative expression levels of glutathione peroxidase 4 mRNA were 1.00±0.11,8.83±0.33,16.03±3.17 and 1.97±0.09;ROS levels were(84.82±0.34),(32.66±0.53),(23.30±0.14)and(64.48±0.33)U·mL^(-1);MDA levels were(6.62±0.05),(2.54±0.02),(1.63±0.04)and(4.97±0.04)nmol·mL^(-1),respectively.Compared with HeLa+SQLE group,the differences of above indexes in HeLa and HeLa-SQLE+mimic-miR-579-3ps were statistically significant(P<0.05).Conclusion miR-579-3p targeting SQLE can promote ferroptosis in cervical cancer HeLa cells,thereby inhibiting the proliferation and invasion of cervical cancer HeLa cells.
