摘  要:目的观察芪蛭通脉颗粒对冠心病模型大鼠基质金属蛋白酶-9(MMP-9)/基质金属蛋白酶抑制剂-1(TIMP-1)的调节作用,研究芪蛭通脉颗粒治疗冠心病的作用机理。方法将Wistar大鼠随机分为芪蛭通脉颗粒低、中、高剂量组,血塞通滴丸组、模型组和空白组,每组10只,共60只。造模成功后,连续用药4周。治疗结束后,ELISA法检测血清MMP-9、TIMP-1和心肌组织MMP-9、TIMP-1水平,检测凝血功能[活化部分凝血酶时间(APTT)、凝血酶原时间(PT)、凝血酶时间(TT)、纤维蛋白原(FIB)]及大鼠肝、肾功能[谷丙转氨酶(ALT)、血清尿素氮(BUN)、血清肌酐(Cr)]。结果与空白组比较,模型组、血塞通滴丸组、芪蛭通脉低剂量组大鼠血清MMP-9水平显著升高(P<0.01);与空白组比较,模型组TIMP-1水平降低(P<0.05)。芪蛭通脉中、高剂量组与模型组比较MMP-9水平降低(P<0.05)。芪蛭通脉中、高剂量组与模型组比较TIMP-1水平显著升高(P<0.01)。各组凝血功能未见异常,模型组ALT、Cr升高(P<0.05)。结论芪蛭通脉颗粒可以降低MMP-9水平,升高TIMP-1水平,可能的作用机制是抑制MMP-9的释放,促进TIMP-1生成,抑制细胞外基质的降解,稳定斑块,延缓动脉粥样硬化进程。未发现低、中、高剂量芪蛭通脉颗粒致凝血功能障碍及肝肾功异常。Objective:To observe the regulation effect of Qizhi Tongmai Granules on MMP-9/TIMP-1 in coro⁃nary heart disease(CAD)model rats and to study the mechanism of CAD treatment.Methods:Wistar rats were randomly divided into Qizhi Tongmai Granules low,medium and high dose groups,Xuesaitong drop pill group,model group and blank group,10 rats in each group,with a total of 60 rats.After successful modeling,treatment was con⁃ducted for 4 weeks.After completion of treatment,serum MMP-9,TIMP-1 and myocardial tissue MMP-9 and TIMP-1 levels were measured by ELISA.Coagulation function(APTT,PT,TT,FIB)and the rats′ALT,BUN,Cr levels were detected.Results:Compared with the blank group,the serum MMP-9 level of rats in the model group,the Xuesaitong drop pill group,and the Qizhi Tongmai Granules low dose group increased significantly(P<0.01).Compared with the blank group,the TIMP-1 level in the model group was reduced(P<0.05).The level of MMP-9 was reduced compared with the model group in the Qizhi Tongmai Granules medium and high dose groups(P<0.05).Compared with the model group,the level of TIMP-1 was significantly increased in the Qizhi Tongmai Granules medium and high dose groups(P<0.01).There were no abnormalities in the coagulation function in each group,and the levels of ALT and Cr increased in the model group(P<0.05).Conclusion:Qizhi Tongmai Gran⁃ules can reduce the level of MMP-9 and increase the level of TIMP-1.The possible mechanism is to inhibit the release of MMP-9,promote the production of TIMP-1,inhibit the degradation of extracellular matrix,stabilize the plate and delay the progression of atherosclerosis.High,medium and low doses of Qizhi Tongmai Granules are not found to cause blood coagulation dysfunction or abnormal liver and kidney function.
