摘 要:目的探讨红参多糖(RGP)对冠状动脉粥样硬化性心脏病(冠心病,CHD)小鼠心肌纤维化的改善作用及其可能作用机制。方法选取40只ApoE-/-雄性小鼠采用高脂饮食法建立CHD小鼠模型,造模成功小鼠随机分为模型组、RGP低、中和高(50、100和200 mg/kg)剂量组,各10只,另将10只C57BL/6J小鼠设为对照组。称取小鼠心脏质量并计算心指数;ELISA法检测各组小鼠血清中丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)和超氧化物岐化酶(SOD)水平;试剂盒检测线粒体复合体Ⅳ、柠檬酸合酶(CS)和顺乌头酸酶(ACO)水平;Masson染色观察各组小鼠心肌胶原纤维沉积情况,并计算胶原容积分数(CVF);RT-PCR和Western印迹法检测小鼠心肌组织中烟酰胺腺嘌呤二核苷酸沉默信息调节因子1(SIRT1)、过氧化物酶体增殖物共激活因子1α(PGC1α)mRNA与蛋白表达情况。结果与模型组比较,RGP低、中和高剂量组小鼠心质量、心指数、MDA和CVF水平降低,GSH-Px、SOD、复合体Ⅳ、CS、ACO、SIRT1和PGC1αmRNA与蛋白表达水平升高(P<0.05)。与RGP低剂量组比较,RGP中和高剂量组小鼠心质量、心指数、MDA和CVF水平降低,GSH-Px、SOD、复合体Ⅳ、CS、ACO、SIRT1和PGC1αmRNA与蛋白表达水平升高(P<0.05)。与RGP中剂量组比较,RGP高剂量组小鼠小鼠心质量、心指数、MDA和CVF水平降低,GSH-Px、SOD、复合体Ⅳ、CS、ACO、SIRT1和PGC1αmRNA与蛋白表达水平升高(P<0.05)。结论RGP可以调节减轻CHD小鼠体内的氧化应激反应,在一定程度上可改善心肌纤维化情况,可能是通过影响SIRT1/PGC1α信号通路,调节心肌线粒体能量代谢发挥作用。Objective The objective of this study was to investigate the potential effects and underlying mechanisms of red ginseng polysaccharide(RGP)in improving myocardial fibrosis in mice with coronary heart disease(CHD).Methods A total of 40 male ApoE-/-mice were selected to establish a CHD mouse model using a high-fat diet.The mice were randomly divided into the following groups:model group,low-dose RGP(50 mg/kg),medium-dose RGP(100 mg/kg),and high-dose RGP(200 mg/kg)groups,each consisting of 10 mice.Additionally,10 C57BL/6J mice were assigned as the control group.The cardiac mass of the mice was measured,and the cardiac index was calculated.Levels of malondialdehyde(MDA),glutathione peroxidase(GSH-Px),and superoxide dismutase(SOD)in the serum of the mice were determined using ELISA.The levels of mitochondrial complex IV,citrate synthase(CS),and aconitase(ACO)were assessed using a kit.Masson staining was performed to observe myocardial collagen fiber deposition,and collagen volume fraction(CVF)was calculated.The mRNA and protein expression of nicotinamide adenine dinucleotide silencing information regulator 1(SIRT1)and peroxisome proliferator-activated receptor gamma coactivator 1-alpha(PGC1α)in mouse myocardial tissue were evaluated using RT-PCR and Western blotting,respectively.Results Compared to the model group,the low,medium,and high-dose RGP groups exhibited decreased levels of heart mass,cardiac index,MDA,and CVF.Conversely,levels of GSH-Px,SOD,mitochondrial complexⅣ,CS,ACO,SIRT1,and PGC1αmRNA and protein expression were increased in the RGP treatment groups(P<0.05).Furthermore,compared to the low-dose RGP group,the medium and high-dose RGP groups showed further reductions in heart mass,cardiac index,MDA,and CVF,while GSH-Px,SOD,mitochondrial complexⅣ,CS,ACO,SIRT1,and PGC1αmRNA and protein expression levels were elevated(P<0.05).Similar trends were observed when comparing the high-dose RGP group to the medium-dose RGP group(P<0.05).Conclusion RGP administration demonstrated the potential to mitigate oxida
