摘  要:为了探究瘦素(LP)和瘦素受体(LEPR)在不同年龄牦牛皱胃中的分布和表达差异,采用免疫组织化学(IHC)、实时荧光定量PCR(qRT-PCR)和Western-blot方法检测不同年龄牦牛皱胃组织中LP和LEPR的定位、mRNA和蛋白的表达情况。IHC结果显示,LP和LEPR蛋白主要分布于皱胃黏膜上皮细胞、固有层胃底腺间的壁细胞、主细胞等部位;qRT-PCR和Western-blot结果显示,LP和LEPR的mRNA和蛋白在初生组、幼年组、成年组、老年组的牦牛皱胃组织均表达,从初生组到老年组表达量依次递增,在老年组皱胃组织的表达量最高,初生组最低。LP和LEPR的mRNA水平在初生组和幼年组之间差异不显著(P>0.05),其余各年龄组之间均差异显著(P<0.05)。LP蛋白表达量在幼年组和成年组之间差异不显著(P>0.05),LEPR蛋白表达量在幼年组和成年组之间差异不显著(P>0.05),其余各年龄组之间均差异显著(P<0.05)。结果显示,LP和LEPR可能调节不同年龄牦牛的皱胃功能,为进一步探究牦牛对高寒低氧的草甸生态环境适应性机制提供基础资料。In order to investigate the distribution and expression differences of leptin(LP)and leptin receptor(LEPR)in abomasum of Bos grunniens of different ages,immunohistochemistry(IHC),quanti-tative real-time PCR(qRT-PCR)and Western-blot were used to detect the localization,mRNA and protein expression of LP and LEPR in abomasum tissues of B.grunniens of different ages.The results of IHC-showed that LP and LEPR proteins were mainly distributed in epithelial cells of abomasum mucosa,pari-etal cells and main cells of lamina propria between gastric fundus glands.The results of qRT-PCR and Wes-tern-blot showed that the mRNA and protein of LP and LEPR were all expressed in the abomasum tissues of newborn group,juvenile group,adult group and eldly group,and the expression levels increased succes-sively from newborn group to eldly group,with the highest expression level in eldly group and the low-est expression level in newborn group.The mRNA levels of LP and LEPR were not significantly different between the newborn group and the juvenile group(P>0.05),but were significantly different among the other age groups(P<0.05).LP protein expression level was not significantly different between juvenile group and adult group(P>0.05),and LEPR protein expression level was not significantly different between juvenile group and adult group(P>0.05),and other age groups were significantly different(P<0.05).The results suggest that LP and LEPR may regulate the function of abomasum in B.grunniens of dif-ferent ages,which provided basic data for further exploring the adaptability mechanism of B.grunniens to alpine cold and low oxygen meadow ecological environment.
