摘 要:通过化学键合透明质酸(HA)和甘草次酸(GA)制备具有CD44受体靶向作用的HA-GA复合物,用以制备聚合物胶束,并采用MTT比色法和酶联免疫吸附分析法等对其制剂学性质和体外抗肝纤维化活性进行评价。结果显示,HA-GA复合物的临界胶束浓度为9.6μg/mL,形成的聚合物胶束粒径为217.0nm,GA含量为18.9%。HA-GA聚合物胶束比GA溶液的细胞毒性强,同时能抑制肝星状细胞分泌转化生长因子β1,表现出了良好的体外抗肝纤维化活性。Hyaluronic acid(HA)and glycyrrhetinic acid(GA)were chemically bonded to prepare the HAGA composite with CD44 receptor targeting property,then the polymeric micelles of HA-GA composite were obtained.The pharmaceutical properties and anti-hepatic fibrosis activity in vitro were evaluated by MTT assay and enzyme linked immunosorbent assay.The results showed that the critical micelle concentration of the HA-GA composite was 9.6μg/mL,the particle size of the polymeric micelles was 217.0 nm,and the GA content was 18.9%.Compared with the GA solution,HA-GA polymeric micelles showed potent cytotoxicity,meanwhile could inhibit the secretion of transforming growth factor β1 by hepatic stellate cells,exhibiting well anti-hepatic fibrosis activity in vitro.
