摘  要:为了解翘嘴鳜(Siniperca chuatsi)miR-133a-5p的时空表达规律以及短期饥饿对其昼夜节律性表达的影响,本研究利用实时荧光定量PCR检测了miR-133a-5p在翘嘴鳜不同组织、不同胚胎发育阶段的表达谱,以及短期饥饿后miR-133a-5p昼夜节律的表达特征。结果显示,miR-133a-5p在红肌中表达最高,在白肌和心肌组织中表达较高,在其他组织中的表达较低;miR-133a-5p在胚胎发育的2细胞期就有较高表达,随着胚胎的发育其表达量逐渐降低,从囊胚晚期开始直至出膜期,其表达均维持在一个较低的水平,且各阶段的表达无显著性差异;miR-133a-5p在正常投喂的翘嘴鳜白肌中的表达没有明显的昼夜节律性,而在饥饿5 d后其表达呈现昼高夜低的节律性。本研究结果表明,miR-133a-5p在翘嘴鳜肌源性组织中高表达,推测其在肌肉组织中有重要作用;miR-133a-5p呈母源性高表达,推测其调控的靶基因可能抑制胚胎早期的发育;miR-133a-5p在饥饿5 d后呈节律性表达,提示其可能通过调节生物节律参与饥饿胁迫下的适应性调控。To understand the spatio-temporal expression pattern of miR-133a-5p and the effect of shortterm fasting on circadian rhythm of its expression in Siniperca chuatsi,real-time fluorescence quantitative PCR was used to detect the miR-133a-5p expression in different tissues and embryonic developmental stages and its circadian rhythm after short-term fasting.The results showed that the expression of miR-133a-5p was highest in slow-twitch muscles,higher in fast-twitch muscles and myocardium,but lower in other tissues.The expression level of miR-133a-5p was high at the two-cell stage during embryonic development,and gradually decreased with the development of the embryo.It maintained at a low level from the late blastula stage to the hatching larvae with no significant difference between each stage.The expression of miR-133a-5p showed no obvious circadian rhythm in fast-twitch muscles of normally fed S.chuatsi,but a circadian rhythm appeared after 5 days of fasting,with high expression during the day and low at night.Taken together,miR-133a-5p was specifically highly expressed in muscle related tissues,suggesting that it may play an important role in the muscular tissue.Its high expression during embryonic development revealed that the target genes regulated by miR-133a-5p may inhibit early embryonic development of S.chuatsi.The rhythmic expression of miR-133a-5p after fasting indicated that miR-133a-5p may be involved in the adaptive regulation of S.chuatsi muscles in response to fasting stress by regulating biological rhythm.
