肝素化聚多巴胺-壳聚糖水凝胶的制备及其抗菌性能

摘  要:通过自由基交联聚合法(FRCP),以聚丙烯酰胺(PAM)为水凝胶基质,引入肝素(Hep)、聚多巴胺(PDA)、羧甲基壳聚糖(CMCS)形成的复合物制备出具有抗菌活性的复合水凝胶。通过紫外分光光度计(UV)、核磁、电子扫描显微镜(SEM)、接触角测试仪等仪器分析其微观形貌及性能,并对水凝胶的亲水性、溶胀率、力学性能、抗菌性能进行探究。结果表明,PAM/Hep-PDA/CMCS水凝胶具有致密多孔、孔径均一的三维交联网络;具有良好的亲水性、力学性能和溶胀性能。抗菌试验表明PAM/Hep-PDA/CMCS水凝胶对金黄色葡萄球菌和绿脓杆菌具有优异的抗菌效果,对金黄色葡萄球菌和绿脓杆菌的杀菌率可高达98.87%、99.84%。该研究为开发具有优异的抗菌活性的水凝胶伤口敷料提供了可行解决方案。Taking polyacrylamide(PAM)as hydrogel matrix,the composite hydrogel of heparin(Hep),polydopamine(PDA)and carboxymethyl chitosan(CMCS)was prepared by free radical crosslinking polymerization(FRCP).Through ultraviolet spectrophotometer(UV),scanning electron microscope(SEM),contact angle tester and other instruments,the micromorphology and properties of hydrogel were analyzed,and the hydrophilicity,swelling rate,mechanical properties,antibacterial properties were explored.The results show that PAM/Hep-PDA/CMCS hydrogels have a three-dimensional cross-linked network with dense porous and uniform pore size.It has good mechanical properties.The antibacterial test shows that PAM/Hep-PDA/CMCS hydrogel has excellent antibacterial effect on Staphylococcus aureus and Pseudomonas aeruginosa.The bactericidal rate of Staphylococcus aureus and Pseudomonas aeruginosa is up to 98.87%and 99.84%.This study provides a reference for the development of hydrogel dressing with excellent antibacterial activity.
