摘 要:以人肺癌A549和H1299细胞为研究对象,探究二氢丹参酮Ⅰ对其自噬的影响.采用qRT-PCR检测不同浓度二氢丹参酮Ⅰ作用人肺癌A549和H1299细胞48 h后自噬相关基因LC3,Beclin-1 mRNA的表达;采用Western Blot检测不同浓度二氢丹参酮Ⅰ作用人肺癌A549和H1299细胞48h后自噬相关基因LC3,Beclin-1蛋白的表达;免疫荧光法检测人肺癌A549和H1299细胞自噬泡.结果表明,经二氢丹参酮Ⅰ处理后,LC3和Beclin-1表达明显高于未加药处理组(P<0.05,P<0.01,P<0.001),LC3Ⅱ/LC3Ⅰ的比值增加(P<0.05,P<0.01,P<0.001);免疫荧光结果显示,经二氢丹参酮Ⅰ处理细胞后,细胞核周围出现集聚的绿色荧光,LC3蛋白集聚形成了自噬泡,表明一定浓度的二氢丹参酮Ⅰ可诱导人肺癌A549和H1299细胞发生自噬.Human lung cancer A549 and H1299 cells were used as research subjects to investigate the impact of dihydrotanshinone I on autophagy.qRT-PCR was employed to measure the expression of autophagy-related genes LC3 and Beclin-1 mRNA in A549 and H1299 cells after 48 hours of treatment with different concentrations of dihydrotanshinone I.Western Blot was used to detect the expression of autophagy related genes LC3 and Beclin-1 protein in human lung cancer A549 and H1299 cells treated with different concentrations of dihydrotanshinoneI for 48 h.Immunofluorescence was performed to detect autophagic vacuoles in A549 and H1299 cells.The results showed that after treatment with dihydrotanshinone I,the expression levels of LC3 and Beclin-1 were significantly higher than those in the untreated group(P<0.05,P<0.01,P<0.001),and the ratio of LC3Ⅱ/LC3Ⅰincreased(P<0.05,P<0.01,P<0.001).Immunofluorescence results revealed that upon dihydrotanshinone I treatment,green fluorescence accumulated around the nucleus,and LC3 protein aggregated to form autophagic vacuoles,indicating that a certain concentration of dihydrotanshinone I can induce autophagy in human lung cancer A549 and H1299 cells.
