摘  要:文章致力于从红树林底泥中分离筛选出能有效抑制细菌群体感应的活性海洋放线菌,为以群体感应效应为靶点的协同抗菌物质研究提供候选菌株。其研究主要采用稀释平板分离法从红树林底泥样品中共分离出159株海洋放线菌,并以紫色色杆菌(Chromobacterium vioLaceum ATCC 12472)为指示菌,采用滤纸片法对其中菌落形态差异较大的61株菌株的发酵产物进行群体感应抑制活性的筛选。实验结果表明,有36株菌株能够抑制紫色色杆菌紫色素的产生,其中菌株HN24的抑制效果最好。结合形态学特征经16S rDNA序列分析,将菌株HN24鉴定为锈赤蜡黄链霉菌(Streptomyces rubiginosohelvolus)。最终研究表明,红树林底泥中放线菌资源丰富,群体感应抑制活性菌株的比例较高,是研究群体感应抑制剂的优良菌源。The paper is dedicated to isolating and screening active marine actinomycetes that can effectively inhibit bacterial Quorum sensing from mangrove sediments,and providing candidate strains for the study of synergistic antibacterial substances targeting Quorum sensing effect.In this study,159 strains of marine actinomycetes were isolated from mangrove sediment samples by dilution plate separation method,and with Chromobacterium violaceum ATCC 12472 as the indicator bacteria,the fermentation products of 61 strains with large differences in colony morphology were screened for Quorum sensing inhibitory activity by filter paper method.The results showed that 36 strains could inhibit the production of purple pigment from Chromobacterium violaceum,among which HN24 had the best inhibitory effect.Based on morphological characteristics and 16S rDNA sequence analysis,the strain HN24 was identified as Streptomyces rubiginosohelvolus.The final research shows that the mangrove sediment is rich in actinomycetes resources,and the proportion of Quorum sensing inhibitory active strains is high,which is an excellent source for studying Quorum sensing inhibitors.
