摘  要:目的 研究槲皮苷对非酒精性脂肪肝大鼠肝纤维化和炎症的影响和机制。方法 将SD大鼠分成对照组、模型组(非酒精性脂肪肝模型)、实验低剂量组(非酒精性脂肪肝模型,10 mg·kg^(-1)槲皮苷)、实验中剂量组(非酒精性脂肪肝模型,20 mg·kg^(-1)槲皮苷)、实验高剂量组(非酒精性脂肪肝模型,40 mg·kg^(-1)槲皮苷)、实验高剂量+佛波醇酯(PMA)组(非酒精性脂肪肝模型,40 mg·kg^(-1)槲皮苷、2μg·kg^(-1)PMA治疗)。用蛋白质印迹法检测大鼠肝组织中p-p65蛋白表达,用苏木精-伊红(HE)染色法评价肝脂肪变性评分和炎症评分,计算肝指数,用比色法检测总胆固醇(TC)、谷丙转氨酶(GPT)、低密度脂蛋白胆固醇(LDL-C)含量,以酶联免疫吸附测定法检测白细胞介素-1β(IL-1β)含量,用Masson染色评价纤维化评分。结果 对照组、模型组、实验低剂量组、实验中剂量组、实验高剂量组、实验高剂量+PMA组肝脂肪变性评分为0、3.33±0.50、2.44±0.53、1.56±0.53、0.89±0.33和2.11±0.33;炎症评分为0、2.78±0.44、2.11±0.33、1.67±0.50、1.11±0.33和2.00±0.50;肝指数为(2.68±0.23)%、(5.06±0.38)%、(4.01±0.47)%、(3.48±0.30)%、(2.99±0.21)%和(3.92±0.15)%;纤维化评分分别为0.22±0.44、3.67±0.71、2.89±0.33、1.89±0.33、1.22±0.44和2.78±0.44。上述指标,模型组与对照组比较,差异均有统计学意义(均P<0.05);实验低剂量组、实验中剂量组、实验高剂量组与模型组比较,差异均有统计学意义(均P<0.05);实验高剂量+PMA组与实验高剂量组比较,差异均有统计学意义(均P<0.05)。结论 槲皮苷通过抑制核因子-κB信号改善非酒精性脂肪肝大鼠肝纤维化和炎症。Objective To study the effect and mechanism of quercitrin on liver fibrosis and inflammation in rats with non-alcoholic fatty liver disease.Methods SD rats were divided into control group,model group(non-alcoholic fatty liver model),experimental low-dose group(non-alcoholic fatty liver model,10 mg·kg^(-1)quercitrin treatment),experimental medium-dose group(non-alcoholic fatty liver disease,20 mg·kg^(-1)quercitrin treatment),experimental high-dose group(non-alcoholic fatty liver model,40 mg·kg^(-1)uercitrin treatment),experimental high-dose+phobolol ester(PMA)group(non-alcoholic fatty liver model,40 mg·kg^(-1)quercitrin and 2μg·kg^(-1)PMA treatment).Western blot was used to detect p-p65 protein expression in rat liver tissue;hematoxylin-eosin(HE)staining was used to evaluate liver steatosis score and inflammation score;liver index was calculated,and colorimetric method was used to detect total cholesterol(TC),glutamic-pyruvic transaminase(GPT),and low density lipoprotein cholesterol(LDL-C).Interleukin-1β(IL-1β)content was detected by enzyme-linked immunosorbent assay;Masson staining was used to evaluate fibrosis score.Results Hepatic steatosis scores in control group,model group,experimental low-dose group,experimental medium-dose group,experimental high-dose group and experimental high-dose+PMA group were 0,3.33±0.50,2.44±0.53,1.56±0.53,0.89±0.33 and 2.11±0.33;inflammation scores were 0,2.78±0.44,2.11±0.33,1.67±0.50,1.11±0.33 and 2.00±0.50;liver indexes were(2.68±0.23)%,(5.06±0.38)%,(4.01±0.47)%,(3.48±0.30)%,(2.99±0.21)%and(3.92±0.15)%;fibrosis scores were 0.22±0.44,3.67±0.71,2.89±0.33,1.89±0.33,1.22±0.44 and 2.78±0.44.Compared with control group,experimental low-dose group,experimental medium-dose group,experimental high-dose group and model group,the difference of the above indicators were statistically significant(all P<0.05);compared between experimental high-dose+PMA group and experimental high-dose group,the differences were statistically significant(all P<0.05).Conclusion Quer
